Purpose: The Paraoxonase in serous is a kind of enzyme which exists in the HDL moleculers, it can hydrolyze Lpos prevents the LDL from oxidizing modification as well as decreases the aggradation of oxidized - type lipids, which can resist the formation of As, that the one of the main mechanisms to anti - As. The further research proves the activity of PON in Familial Heterotyped Hyper-cholesterdemia and diabete patients is relatively lower , comparing with the contrast group. Because these two kinds patients is liable to be attached by As, we can induce PON has the closely relation with the development of As. The diabete patients that affect the complications such as coronary heart disease, ische-mic encephic blood vessel disease, limb gangrene and so on are obviously higher than the non - diabete people. The Doppler Ultrasound Equipment can detect the lower blood vessel lesion earlier. So we use this technology to detect the degree of the As in lower limbs and use the photometer to measure the absorption of 270nm, which can detect the affected activity of Paraoxonase in the serous of diabete patients with the complecation of Atherosclerosis, research the effect of PON to the diabete patients with the complecation of Atherosclerosis,Methrial and MethodsSEROUS ( healthy people) ; the inspection department of the first attached hospital of CHINA MEDCINE UNIVERSITY. The diabete patients coming from our hospital during 2002 to 2003, accords the diagnosed criterion of ADA. The activity of PON is measured by photometer using the 270nm absorption, Buffer solution 2. 6ml ( CaC12 2mol/L, Tris/HCL 0. 1mmol/L PH = 8. 0), 5mmol/l ACETIC, heating up for 20mins in 25℃ , adding 1:10 serous sample 50ml,after90s , adding 0. 5mol/l EDTA to end the reaction. Measuring the absorption of the solution in 270nm. Meantime building the contrast groups of serous and subsides. All the subsides must be newly confected every time. The formula of activity of the enzyme is: PON activity kv/L = Axl03 xfx FV/t x S x VL x eResults1 the two group of diabetes compared with contrst group is detected the Glucose , Glycerin, Cholesterol, HDL, LDL.2 the two group of diabetes compared with contrst group is detected the activity of PON in serous.3 the activity of PON in diabete patients without the complecation of Atherosclerosis is lower than the normal contrast group ( P < 0. 01 ) , the activity of PON in diabete patients with the complecation of Atherosclerosis is lower than the diabete patients without the complecation of Atherosclerosis ( P <0. 05) , the difference is obvious.4 the activity of PON in normal serous is irrelatively with TG, TC, HDL - C and LDL-C and HDL-C, ApoB (P>0.01).5 the Km of PON ; 0. 87mmol/l, is close to the result of reference (Km = (0.85 0.04)moJ/l).DissusionThe Paraoxonase in serous is a kind of enzyme which exists in the HDL moleculers, it can hydrolyze Lpos and phosphriside. It is more and more concerned for the action of resisting the As. The As is one of the chronic complications of diabetes, which mainly invades the aorta A, coronary A, encephic A, renal A,and peripheral A. The peripheral As usually occurs in the lower limbs. The results from the Doppler Ultrasound equipment shows that the activity of PON in the diabete patients without As in lower limbs is lower than the normal contrast group, the activity in the diabetes with As in lower limbs is lower than the diabetes without As in the lower limbs. All these prove the activity of PON isrelative with the diabetes and the As.Michael and so on proved that the decreasing activity of the PON in serous is relative to IDDM and FH, large experiments have proved that the PON is closely relative to HDL, the anti -lipid oxidizing action of HDL is very clear, PON the composition of HDL, directly involved with hydrolyzing the peroxide and prevent the aggregation of peroxide to the LDL, as a result, the activity of PON can reflect the ability of the anti - lipid oxidizing action of HDL, the decreasing activity of PON also...
