Aim: To investigate the roles of CD 14 and Scavenger receptor(SR) in the Bile Duct emphraxis-induced Hepatic injury, and the influence of glycine to CD14 and SR's expression.Methods: A mode of Bile Duct emphraxis was employed. The mice were randomly divided into control, Bile Duct emphraxis 7, 14 , 28days and glycine therapy 7, 14 , 28days groups. With the model of Bile Duct emphraxis in mice, the mice were caudal vein injected with glycine. The dose of glycine is 100mg/kg, the concentration of glycine solution is 1.5%. Themice were harvested after Bile Duct emphraxis by 7, 14, 28days respectively. Plasma LPS, ALT, AST, TB and liver Homogenate TNF-a were observed. CD 14 and SR were observed by Immunohistochemistry on frozen.Results: Experimental results indicated that : (1), With the Bile Duct emphraxis developing plasma LPS, ALT, AST, TBand liver Homogenate TNF-a were all increase and showed positive correlation among them. The expression level of SR was decreased and CD 14 was increase at the same time and showed negative correlation among them. (2) After injected with glycine, plasma LPS, ALT, AST, TB and liver Homogenate TNF-a were all decrease. And the expression of SR was increased and CD 14 was decreased.Conclusions: (1) LPS can decrease the expression of SR, increase the expression of CD 14. Through these two ways LPS can weaken the Kupffer cell's ability of cleaning LPS and enhance the KC's activing, therefore active the inner signal transduction system, release lots of cytokine and lead to more serious Hepatic injury, formatting the second bit to liver.(2) As a inhibitor of Cl -passage, glycine can uptake the expression of SR and enhance the Kupffer cell's ability of cleaning LPS, At the same time, restrain the expression of CD 14 and repression the LPS's biology effect. This experiment indicates that glycine can therapy IETM and improve Liver function effectively. It is clear that additional investigation is necessary to fully appreciate the role of the glycine in the expression of CD 14 and SR.
