Objective To investigate the effects and mechanism of Panax Notoginseng Saponins (PN6S) on the rat liver during cold preservation, and screen optimal concentration of PNGS on the rat liver. Methods Using isolated perfusion of ratliver model(IPRL) , DMEM solution and Lactate Ringer' s (LR) solution containing PNGS with different doses and Verapamil(Ver) 20 mg respectively, Fura-2 method was used to measured the intracellar calcium of the 2 hours cold preservation;the liver function, free radical,energy substance and bile production, hepatocellular apoptosis and Bcl-2 protein expression, morphologyical analysis were measured after 24 hours cold preservation and 30 minutes reperfusion to compare with the protective effects of PNGS and Ver on rat liver during cold preservation- reperfusion. Results the contents of intracellar calcium of the rat hepatocytes, MDA, SOD, AST, ALT, LDH, ATP, AEC, TNA and bile production, hepatocellal apoptosis and Bcl-2 protein positivly express ion were significantly better than the control group (P<0. 01) . Dur i ng 200-600mg, the protect i ve effects of the PNGS were correlated with the dosage in the LR solution and manifest dosage-dependence (P <0.01), 600-1000 mg were not significant (P >0. 05). The protective effects of the Ver is only correspondent with PNGS 200-400 mg(P<0. 01). Conclusions PNGS could obviously improve the preservation effects of donor liver. The mechanism might be that it could inhibit calcium overload, improve the energy metabolism , play a role against free radical injury and hepatocellular apoptosis. The protective effects of the PNGS was better than Ver, it might be correlate with different mechanisms of inhibit ion of calcium over load and free radical injury.
