Objective: To inspect the expression of mdm2 and p53 in lung cancer of Gejiu miners and non-miners of Yunnan, and investigate the role of alterations of mdm2 and p53 in the tumorigenesis of lung cancer of Gejiu miners.Methods: Immunohistochemical technique (S-P) method and in situ hybridization were used to study the expression of mdm2 in 44 cases of lung cancer of Gejiu Tin miners , 42 cases of lung cancer of non-miners of Yunnan and 15 cases of normal lung tissues. The immunohistochemical result of MDM2 protein was compared with the immunohistochemical result of p53 protein which was performed in early study in 15 cases of lung cancer of Gejiu Tin miners.Results: There was difference between the positive degree of the expression of MDM2 protein in lung cancer of Gejiu miners and non-miners of Yunnan (P<0.05). But MDM2 protein was not found in 15 cases of normal lung tissues, there was difference between lung cancer and normal lung tissue (P<0.01) . There was difference between the positive rate of the expression of mdm2mRNA in lung cancer of Gejiu miners and non-miners of Yunnan (P<0.10), and there was difference between lung cancer and normal lung tissue (P<0.01) . There was correlation between the expression of MDM2 protein and mdm2mRNA (P<0.01). There was correlation between the expression of MDM2 protein and the mutation of p53 (P<0.01) .Conclusion: The expression of MDM2 protein in lung cancer of Gejiu miners was stronger than that of non-miners. It indicated that the tumorigenesis of lung cancer was different between Gejiu miners and other regions according to different pathogeny. Over-expression of MDM2 protein in lung cancer may be caused by enhanced transcription. There may be cooperation between the expression of MDM2 protein and the mutation of p53 in the tumorigenesis of lung cancer of Gejiu miners. |
