| Endothelia (ET) and calcctonin gene -related peptide (CGRP) are respectively the strongest biologic active multi - peptides to contract blood vessels and diastole blood vessels in vivo. In vivo, they widely distribute in central, peripheral nerve system and cardiovascular system and some organs. These two kinds of multi - peptides'regulating effect on blood vessel tension may have major influences on essential hypertension ( EH). The research determines the contents of ET and CGRP in EH patients'blood plasma and oral Valsartans influence on these two kinds of multi - peptides are observed.Methods1. Objectives30 in - patients who are diagnosed as EH under the circumstance of no and high blood pressure medicines taken with systolic pressure > 140mmHg and /or diastolic pressure > 90mmHg according to high blood pressure diagnosis standard, from Feb. 2000 to Aug. 2000, in accordance with the definition of 1999 2 (WHO/ISH), male 17 cases, female 13 cases, average age 54. 5 ±10.3 (37-78). Amongothers 7 cases at high blood pressure level 1, 16 cases at level 2, and 7 cases at level 3. Through routine urine test, kidney function, blood electrolyte, eyeground, electrocardiogram, and B ultrasonic examinations , such factors as secondary hypertension, serious incompetence of liver, kidney functions are excluded.In the control group 20 cases, male 14 cases, female 6 cases, average age 37.8 ±11.8(18~56),are those who take medical examinations and are volunteers. They are healthy, have normal blood pressure, and excluded from other medical illnesses.2. MethodsAll patients stop to take any medicines to reduce blood pressure within one week at least before the test. After getting up in the morning at 7 o'clock test the patients'blood pressure through right upper limb with sitting position. After testing blood pressure, take 3 ml vein blood. Then the patients take Valsartan 80mg orally everyday, once a day, total 14 days. 3 hours after taking the medicine in the morning of 14th day, measure blood pressure and take blood with the same method, measure blood pressure three times continuously and choose an average value.3. Blood plasma ET and CGRP level determinationTake radio - immunity analysis method to directly determine blood plasma ET and CGRP level. Inject peripheral vein blood 3 ml in a plastic tube fitted with anticoagulant. After mixed evenly, whole blood centrifuge (3000r/min, -4℃, 10 minutes) separate blood plasma. Put the specimen in a refrigerator under - 70℃ to save for assay.4. Statistics treatmentThe data is expressed by average deviation and standard devia-tion. T test and linear test for correlation.Results1. ET and CGRP levels in blood plasma of EH patientsET and CGRP levels in blood plasma of EH group are respectively 88.6 ± 10. 7 pg/ml and 24. 3 ±17.4 pg/ml, those of the control group are respectively 46. 4 ± 8. 4 pg/ml and 51. 2 ±20. 6pg/ml. Compared with thh control group the differences of the two indexes are all of evidence. ( p <0.001 without exception).2. the relation between EH patients blood pressure and blood plasma ET and CGRP levelRelevant analysis of EH patients'systolic pressure, diastolic pressure , and blood plasma ET and CGRP level shows that there is not evident correlation between systolic pressure and ET, diastolic pressure and ET present positive correlation ( r = 0. 536, p < 0005 ). There is not evident correlation between systolic pressure, diastolic pressure and CGRP. ET and CGRP present negative correlation. ( r = -0.3105, t=2.145, p<0.05).In the test it is obvious that after 14 days taking Valsartan, EH patients'blood pressure reduce evidently. ET level in blood plasma reduces evidently, and CGRP level rise obviously.Discussions1. changes of ET and CGRP level in blood plasma of EH patientsET levels in blood plasma of his group of EH patients is obviouslyhigher than those of the control group, and present positive correlationwith diastolic pressure of EH patients.
