Subject: The aim was to study the effects and mechanism of Naoshangtai(NST) for vascular dementia(VD) in Wistar rats.Method: In this study, a rat model made by Pulsinelli's 4-vessel occlusion method was used. The rats were randomized into control (sham injury) group, vascular dementia (VD) group, vascular dementia rats treated with Naoshangtai(NST) group and Naozhenning(NZN) group. On the week 1, 2, 4, 6, and 8, the correct rate of active avoidance response (AAR) was detected. On the week 8, P300 wave was recorded by brain evoked potential machine. On the week 1, 2, 4, 6, and 8, five animals of each group were sacrificed and the samples were obtained. Electron microscope was used to observe the morphology of neuron in hippocampus. The hippocampus were then stained with immunohistochemistry to detect the expression of calbindin(CaB) and synaptophysin (SY).Results:1. There were significantly changes of the correct of active avoidance response, delay of latency of P300 wave, and damage of neurons in hippocampus on VD rats. The expressions of synaptophysin and calbindin were obviously reduced in hippocampus.2. Compared with VD group, the rats of NST group and NZN group had significantly improvements of correct rate of AAR, latency of P300, and damage of neurons in hippocampus.3. On the week 6 and 8, it were observed that the expressions of SY and CaB had significantly increased in hippocampus of NST group and NZN group. Conclusion:1. The Pulsinelli's 4-vesseloccIusion was a better method for vascular dementia on rats. The dysfunction of learning and memory of the rats involve in the damage of neurons and down-regulation of expressions of SY and CaB in hippocampus.2. NST and NZN could improve the ability of learning and memory in VD rats. It may be their mechanism that NST and NZN could improve the neuron ischemic damage and up-regulate the expressions of SY and CaB in hippocampus in VD rats. It was more effective treated with NST than NZN.
