Objective:To study the effects of exogenous tetramethyl. Pyrazine (IMP) and prostaglandinEi (PGEi) on cerebrocellular apoptosis of hypoxic-ischemic brain damage (HIBD) in neonatal rats,and on either the SOD orNO level in brain tissue. Methods:Select 7-day old newborn Wistar rats to establish HIE mocid,and intraperitoneally and subcutaneous injected TMP and PGEi then the rats were killed after hypoxia and ischemia for another 48 hours.The number of cerebrocellular apoptosis,apoptotic rate and apoptotic dense of rats were detected after the brain tissue were stained by HE and TUNEL. For parts of rats,taken cerebral cortex of arteria carotis ligation side and made them into homogenate to detect SOD and NO levels in brain tissue,compare them into normal control,HIE group,normal saline groups. Results:The number of cerebrocellular apoptosis,apoptotic rate and apopototic dense in HIE group were much higher than that in normal control group (p<0.01),the value in TMP group.PGEi group and.TMP+PGE1 group was much lower than that in HIE group and normal saline group (p<0.01,p<0.01,p<0.01),the value in TMP+PGEi group was much lower than that in TMP group and PGE1 group(p<0.05). SOD lever in brain of HIE group were lower,and NO lever were higher,they were significantly different with that of normal control group (p<0.05),SOD lever ofIMP group,PGE1 group,TMP+PGEi group were much higher,and NO lever were lower,they were significantly different with that of HIE group and normal saline group (p<0.05). When TMP and PGEt used together,the detected SOD( NO )level were much higher(and lower) than that when TMP andPGE1 used separately groups(p<0.05).Conclusion:(1)As a Calcium antagonist,TMP can decrease cerebrocellular apoptosis and promote the increase of SOD in brain tissue with hypoxic-ischemic damage,reduce the level of free radical,So it have prevention and cure effect on HIE.(2)PGEi can pass through the blood-cerebral barrier and inhibit cerebrocellular apoptosis,can promote the level of SOD and NO in cerebral cortex,So it have prevention and cure effect on HIE (3)when TMP and PGE1 treated together,the effect was better than the two treated separately,they are coordinate and complement each other and can enhance curative effect.
