| Objective: To investigate the distribution of TCR V 13 subfamily T clonal cells of patients with acute myelogenous leukemia M2a subtype (AML-M2a) and cytotoxicity of T cells induced by AMLM2a cells in vitro for the possibility of obtaining one kind of ideal cells to treat minimal residual disease(MRD). Methods: Complementarily determining region 3(CDR3) size of I cell, antigen receptor(TCR) 24 variable region gene was analyzed in peripheral blood lymphocytes(PBL) samples from 17 AML M2a cases using reverse transcriptase-polymerase chain reaction(RT-PCR) and genescan techniques, to identify the distribution and clonality of TCR V P subfamily T cells. In addition, mixed lymphocyte and tumor culture(MLTC), was used to investigate the ability of AMLM2a cells to induce T clonal cells from the AML.M2a patients and normal individual; The phenotype and cytotoxicity were analyzed by indirect immunofluorescence and MTT methods, respectively. Results: All 24 TCR V P subfamily T cells were detected in normal cases, whereas only 3-13 V 13 subfamily T cells were found in patients with AMLM2a. V 1313 subfamily was expressed mostly in dominant V P subfamilies, V 13 2 and V f33 were in the next place at the same time. Individual othemess was the general characteristic of dominant TCR V 13 subfamily T cells, however. Clonal T cells could be found in some dominant TCR V P subfamily T cells in every patient. The similary distribution of dominant TCR V 13 subfamily T cells were found in T cells both from patients and which induced by AMbM2a cells in vitro MLTC, difference of distribution was appeared when auto-T-cell or allo-T-cell were induced by the same AMLM2a cells at the same time. Some of dominant V P subfamily I cells displayed clonal expansion. Compared with LAK cell, most of T cells from MLTC are CD3 CD8T cells with higher specificity and cytoxicity toward the induced cell, AMLM2a cell, but not HL6O cell line and K562 cell line.Conclusion: Clonal expansion and dominance were the character of ICR V 13 1 cells in AMLM2a patients. I clonal cells with specific cytoxicity can be obtained from PBL of both AML M2a patients and normal individual induced by AML.M2a cell, which may be one kind of desired cells clonality to treat MRD. |
