Objective The purpose of this paper was to study the effects of bone marrow stromal cells(BMSC) cultured in vitro on repairing the bone defects, and to provide experimental basis of repairing the bone defects and bone nonunion With cultured BMSC in clinic. Methods Numbered the experimental rabbits. Two milliliter bone marrow was obtained by percutaneous aspiration from the medullaiy cavity of the femur through the greater trochanter and cultured in five 20m1-culture-flasks for 14 days. (4 X 106 cells per flask).The morphology of the cultured BMSC was observed with light microscopy everyday. On the eleventh day of culture,one and half centimeters long periosteum-bone defects of the two radii of every experimental rabbit were obtained by surgical operation. Three days after the operation, the bone defects of the experimental side was autotransplanted with 1 X 108 BMSC cultured in vitro for 14 days, and the control side were injected with the same volume culture medium as the bone marrow stromal cells suspension. The X ray examination was done at the end of first, second, third, and fourth week after transplantation.The Lane-sandhu evaluating method was used to evaluate the quantity of the callus formed in bone defects which t displayed on radiograph.
