PURPOSES: To examine the expression of M30 CytoDEATH and p53 in non-small cell lung cancer (NSCLC) , and investigate the tumorigenicity of lung cancer by learning the relationship between M30 or p53 immunoreactivity and clinicopathological parameters in NSCLC. METHODS: The expression of oncosuppressor gene p53 and monoclonal antibody M30 CytoDEATH in 62 archival specimens of patients with resected NSCLC (included 29 cases squamous cell carcinoma and 33 cases adenocarcinoma) and 10 cases of control group were examined by adopting immunohistochemistry assay (LSAB). Moreover, the expression of p53 immunoreactivity with positive unit (PU) was quantitatively evaluated by a micro-image analyzing instrument (Leica Q500MC). RESULTS: 1. The expression of p53 in NSCLC and its relationship with climcopathological parameters: (1) In 62 NSCLC, p53 immunoreactivity was detected in 15 of 29 squamous cell carcinoma (51.72%), 15 of 33 adenocarcinoma (45.45%), 30 of 62 NSCLC (48.39%). In 10 control cases, no expression of p53 was found (0.00%). There were significant differences between these groups (qualitative analysis p53 (? 1+) p=O. 004; quantitative analysis p53(PU): p~0. 000). (2) No relationship between the expression of p53 and T~NM staging or lymphonode invasion or distant metastasis was found in NSCLC. (3) The expresson of p53 had a positive con-relation with differentiated degree of lung adenocarcinoma, qualitative analysis p53 (- 7+) p=O.OOS; quantitative analysis P53(PU): p=0.035. It suggested that the expression of p53 in well- differentiated adenocarcinoma was lower than the moderately- and lowly- differentiated ones. 2. The expression of M30 CytoDEATH in NSCLC and its relationship with clinicopathological parameters: (1) In 62 cases NSCLC, the apoptotic index (AI%) of M30 is 1.10%, and in 29 cases squamous cell carcinoma is 0.95%, and in 33 cases adenocarcinoma is 1.24%. In 10 control cases, the AI% is 1.06%. There is not significant difference among these groups. (2) No relationship between M30 expression and metastasis was found in the NSCLC group, the squamous cell carcinoma group and the adenocarcinoma group. (3) No relationship between M30 expression and TNM staging was found in the NSCLC group and the squamous cell carcinoma group, but there was significant difference found between the expression of M30 and TNM staging (p=O.O42) . It suggested that AI% of the higher TNM staging was higher than the lower ones in lung adenocarcinoma group. 3. According to the results of Pearson's correlation analysis: (1) No linear con-elation was found between the expression of M30 and p53 in NSCLC. (2) We found positive linear con-elation among the expression of p53 (-7+) , p53 (5 degrees) and p53 (PU) (p=O.000). CONCLUSIONS: 1. The significant overexpression of p53 emerged in NSCLC suggested that mutated type gene p53 possibly related to the developing mechanism of NSCLC. 2. The expression of p53 and M30 suggested that the developing mechanism of NSCLC possibly related to cell excessive proliferation and insufficient apoptosis. 3.
