Damas1869 was used as materials due to its anti-waterlogging and grafting compatibility with peach. A stable and high efficient micropropagation system was established, different factors that effect adventitious shoot regeneration were also studied in order to establish efficient plum regeneration system. The results were as follows:Firstly, micropropagation in Vitro.①Experiment was carried out using the shoots in winter and spring of Damas1869 as material. The results showed that the tender shoots from cutting in water during winter was the best explants. The rate of survival was 64.0%. The best method for disinfectant was 75% alcohol (washed for 10s) and 20%NaClO (washed for 8m).②The optimal medium for shoot generation was MS+6-BA1.0 mg/L+NAA0.5mg/L+ 20.0g/L sugar + 5.0g/L agar with 14h/8h light period, 2000light intensity and 25℃temperature. The multiplication coefficient could be reached 7.1.③The optimal medium for rooting was 1/2MS + IBA2.0mg/ L + 20.0g/L sugar + 5.0g/L agar + 0.05% activated charcoal, , the rooting rate was 87.8%, and the mean number of roots was 3.9.④It is better to cultivate the in vitro buds two times in low hormone medium continuously before planting. Turfy soil and perlite and vermiculite (1:1:1) was useful for transplantation with plants higher than 4cm. Keeping humidity within a week after transplantation was also useful for the surviva1.The surviva1 rate would be reach 90%. Secondly, leaves regeneration①Experiment was carried out using the young leaves in vitro for studying the effects of the different basic culture media, the contents of TDZ, 6-BA, IBA and NAA, agar on the adventitious shoots regeneration.The results showed that F14 medium was the optimal basic culture medium for the adventitious shoots regeneration and better than MS and WPM medium.②The optimal hormone contents were TDZ 4.0mg/L + IBA0.5 mg/L.③Regeneration medium with lower agar content (4.5g/L) was an effective method to improve adventitious shoot regeneration with the leaf adaxial side touching media and five days dark pretreatment. The regeneration rate was 73.5% and 3 to 5 shoots per leaf.
