Flower industry is a part of the modern agriculture. With the improvement of the progress of the society and living standards of the people, the market demand of the flowers grows and flowers have already become the indispensable consumer goods in social life and daily life. The market potential is expanding constantly. Flowers become the important goods that primary industry (planting ) entered the tertiary industry directly gradually, and have very wide market prospects. Plant tissues culture technology offer the new way for the fact that the flowers are developed and improved with the properties. Compared with traditional vegetative propagation method, plant tissues culture technology has advantage of uniqueness.lt can breed some improved seeds and get rid of poison to go on fast, and become possible to produce while being factory to make.Tissues culture of the horticulture plant has gone through a history in the halfth century. Especially since the sixties of the 20th century, it develops rapidly in each field of horticulture. As a new technology, tissues culture can be used in hereditary breeding , fast reproduction extensively, getting rid of poison,and kinds of variety resource keeping and exchanging etc..The past 20 years, plant tissues culture technology to develop and improve constantly breed the flowers not only have made the multiplication speed increase greatly but also obtained the new vegetative propagation way . It offer the new thinking for problems of solving sex degradate of precious flowers kinds and beeding .Gladiolus hybridus is the perennial herbaceous corm plant of Iridaceae Gladiolus. Gladiolus is one of the common flowers of the courtyard of the world. In recent years, The market demand of Gladiolus increases constantly with the development of social economy. It is one of the main cutting flowers varieties in the world at present, and its sales amount occupies the first place in the cormflowers. Gladiolus is mainly breed by way of dividing cormlets in production, and adopt seeds to breed in crossbreeding,but the rate of proliferation is low and the long-term vegetative propagation is apt to be infected with the virus and caused degradating so that influence output and quality of Gladiolus. These disadvantage can be remedy by tissue culture. Tissue culture can improve the multiple of proliferating effectively , shorten proliferating cycle , and can make flowers grow for achieving the goal of supply of the flower in anniversary. In addition improved seeds can be update and no virus plant be obtained by ,so variety degradate caused by long-term vegetative propagation can be solve. Tissue culture has better actual production and value. So tissue culture of Gladiolus is paid attention to extremely.The development of molecular biology and gene engineering has offered a brand-new breeding way for improving and decorating the properties of the flowers, people can channel the function gene into the tissues and cells of the specific variety purposefully snd pointedly , in order to get the transgenic plant with goal properties. Combine genetic engineering with traditional breeding means, in the research in the future, cultivate large quantities of various colours, strong resistance, different properties and different purposeful flowers varietying.In the development of plant genetic engineering, the study on transformation mediated by Agrobacterium tumefaciens is clear and use successful. The transformation system mediated by Agrobacterium tumefaciens is not only mature transformation method of plant in theory and technology at present, but also the high rate of transformation and the best hereditary stability of transforming plant.Gladiolus is monocotyledon of Iridaceae. Although the transformation means mediated by Agrobacterium tumefaciens has a lot of difficulty, already there are reports that can be transformed. The high-frequency regeneration system is an important condition succeeded in transforming.The explants are dormant cormlets of Gladiolus in the study. Various kinds of influence factors were systematically studied in the course of tissue culture. The research that explored optimum terms of Gladiolus in rapid propagation and set up high-frequency regeneration system established a solid foundation of wide application for obtaining new variety of Gladiolus applying transgenic method. Studing the formation of Gladiolus comlets in vitro can be applied to industrialization of Gladiolus.Main research work and result:(1) The optimization of the condition in tissue cultureFor this study, the slices of dormant cormlets with buds and no bud were adopted as explants for tissue culture.
