Function Analysis Of Put-R40g3 Gene In Puccinellia Tenuiflora Under Abiotic Stress

At the present time,growing area of salina played bad impact in agriculture and economy. So it's pretty important to exploit and reuse this kind of resource.This thesis is based on that situation and takes Puccinellia tenuiflora as our experiment material.Put-R40g3 gene(accession number:AB465547) was isolated from Puccinellia tenuiflora.The ORF of Put-R40g3 gene is 869 bp which encoded 195 amino acids and showed high similarity to R40g3 protein in Triticum aestivum,Oryza sativa and Arabidopsis thaliana. RT-PCR analyses showed that the expression of Put-R40g3 gene in root and leaf of P.tenuiflora was induced by several abiotic stresses from salts,drought and ABA treatment. This result indicated that Put-R40g3 gene has close relationship with abiotic stresses.Yeast cell is used as model to study the location of Put-R40g3 protein and tolerance when Put-R40g3 gene expressed in yeast cell.The Put-R40g3 protein was localized in cytoplasm by using the green fluorescent protein(GFP) marker.At the same time,transgenic yeast expressing Put-R40g3 gene increased the resistance to salt,drought,oxidative stresses and ABA treatment.In several transgentic Arabidopsis thalianas,which express Put-R40g3 gene show better in condition of radication when under salt and drought stress.Otherwise,transgentic Arabidopsis thalianas show different with wild type in configuration.This indicated that Put-R40g3 protein is related to growth.Above all,this thesis only shows brief analysis in character of Put-R40g3 gene.About the function and character of Put-R40g3 protein and what kind of role that Put-R40g3 protein plays in abiotic resistance need more research.
