Genetic variation of different populations of Red lion-head goldfish,Variegated lion-head goldfish and Red crest tiger-head goldfish were carried out by morphological variation, SSR and non-special immunity.The morphological and truss network characters of different populations of Red lion-head goldfish, Variegated lion-head goldfish and Red crest tiger-head goldfish were studied by three multivariate analysis (cluster analysis, discriminate analysis and principal component analysis). The result shows that Suzhou population has a close relationship with Nantong population, and Hangzhou population is far from other two populations of three goldfishes.Population genetic variability and genetic structure of three populations of Red lion-head goldfish, Variegated lion-head goldfish and Red crest tiger-head goldfish were analyzed by using 10 microsatellite loci. The results shows that the average heterozygosity of three populations of Red lion-head goldfish ranges from 0.1050 to 0.1985, which is 0.1218 for Suzhou population, 0.1985 for Nantong population, and 0.1050 for Hangzhou population. The genetic similarity index between Suzhou population and Nantong population is the highest (0.9420), and between Nantong population and Hangzhou population is the lowest (0.8072). The average heterozygosity of three populations of Variegated lion-head goldfish ranges from 0.2627 to 0.3215, which is 0.3215 for Suzhou population, 0.2627 for Nantong population, and 0.3104 for Hangzhou population. The genetic similarity index between Suzhou population and Nantong population is the highest (0.9513), and the lowest between Nantong population and Hangzhou population (0.8550). The average heterozygosity of three populations of Red crest tiger-head goldfish goldfish ranges from 0.1670 to 0.3007, which is 0.3007 for Suzhou population, 0.2763 for Nantong population, and 0.1670 for Hangzhou population. The genetic similarity index between Hangzhou population and Nantong population is the highest (0.9413), and the lowest between Suzhou population and Hangzhou population (0.6864). Comparing the percentage of erythrocyte active rosette(EaR) of T lymphocyte, the activity of leukocyte phagocyte and LZM of different populations of Red lion-head goldfish, Variegated lion-head goldfish and Red crest tiger-head goldfish in the condition of water with 100mg/L NH4Cl and that without NH4Cl, the results of Red lion-head goldfish show that the percentage of erythrocyte active rosette(EaR) of T lymphocyte ranges from 4.57% to 4.92%, the activity of leukocyte phagocyte ranges from 9.32% to 9.67%,LZM ranges from 0.012 to 0.014; the results of Variegated lion-head goldfish show that the percentage of erythrocyte active rosette(EaR) of T lymphocyte ranges from 4.17% to 4.42%, the activity of leukocyte phagocyte ranges from 8.13% to 8.37%,LZM ranges from 0.011 to 0.013; the results of Red crest tiger-head goldfish show that the percentage of erythrocyte active rosette(EaR) of T lymphocyte ranges from 4.02% to 4.32%, the activity of leukocyte phagocyte ranges from 8.03% to 8.27%,LZM ranges from 0.010 to 0.013. There are no significant differences among the different populations of three goldfishes.
