Study On The Culture Conditions Of Suillus Luteus

Suillus luteus is a superior mycorrhiza fungi,but its application is still limted for the slow growth in liquid culture.In this paper,the effect of culture medium, innoculation time,adding substance,pH,carbon resource concentration and nitrogen resoure concentration on hyphal growth were investigated,was proceeded to determine influence factor of Suillus luteus growth and establish a effective culture systerm.The result showed that,the optimum culture media was Pach.The optimum innoculation time was 6-8d.Mushroom extracts promoted the hypha growth of Suillus luteus,and the Flammulina velutipes extracts was the most useful. The citric aicd-K2HPO4 buffer was optimum for the hyphal growth,and the optimum buffer concentration was 30-40%.Suillus luteus grew well in pH4.5.Under different pH conditions,the optimum carbon resource concentration was increased with the pH increasing,and the optimum nitrogen concentration have a little change.Under solid culture condition,the optimum pH and carbon resource concentration's combination for hypha growth was pH4.5-glucose concentration0.1mol/L,the optimum pH and nitrogen concentration's combination was pH4.5-tartaric ammonium concentration0.014mol/L. Under liquid culture condition,the optimum pH and carbon resource concentration's combination for hypha growth was pH4.5-glucose concentration0.1mol/L,the optimum pH and nitrogen concentration's combination was pH4.5-tartaric ammonium concentration 0.007mol/L.
