| Transfer factor (TF), which are complex of small peptide and oligonucleotide , are a kind of small active molecule extracted from lymphocytes. TF can transfer donor's cell mediated immunity to receptor's cell mediated immunity. Particularly unspecific TF can enhance receptor's natural immunity and specific immunity, which makes TF being applied at large in treatment and prevention of animal and human diseases. Canine parvovirus (CPV) is a kind of acute disease, which is caused by Canine parvovirus. The characteristic clinical symptoms of CPV are vomiting, bloody enteritis and serious leucopenia, which also cause acute cardiac muscle inflammation. The symptoms are similar to the feline panleukopenia. CPV is one of the most infectious diseases in our country.That TF can markedly enhance canine cellular mediated immunity has been borne out at the prophase test. The further research was done to study whether swine and canine TF can as vaccine diluents simplify immunization methods and procedures. The effects of swine and canine TF to proliferate of CPV in MDCK cells in vitro were investigated. Using two different methods vaccinate dog with swine TF or canine TF and vaccines to study on the effects of swine and canine TF to humoral immunity of dog. The main results are as following:CPV was proliferated in MDCK Cells to offer antigen for the following research. The conditions of hemagglutination test are optimized to detect CPV. The best conditions to detect CPV are : 50μL , 1 % glutaraldehyde-fixed RBC , 0.01 M PH6.8 PBS as thinner, acting 60 minute in 4℃. The titer of hemagglutination test of vaccinate diluents is 26, when the titer of the cell cultures of vaccinate diluents is 211. The results show that the vaccine is attenuated CPV Vaccine.Different dosage of swine or canine TF adding to MDCK cells can singly induce cytopathic effect (CPE), the CPE appearing time is deferred following the reduce of swine or canine TF. The effects on MDCK cells of swine TF are more obvious than canine TF's. Adding swine or canine TF to MDCK absorbed CPV, the CPE appearing time is earlier than TF or vaccine adding to MDCK cells only. TF made the proliferation of canine parvovirus on MDCK weakness. When applied same dosage, the effects of swine TF is more obvious than canine TF's.Injecting 3 mL swine TF or canine TF and vaccine to dog, collecting blood every 5 days, using hemagglutination inhibition test to detect, and the titer of different group is similar. Using swine TF or canine TF as thinner of vaccine, the titer of different group has no difference.The results showed that TF could be used as thinner of vaccine. However, TF's applied dosage needs further research. |
