The aim of this experimentation was to establish a rapid technique system of micropropagation.The buds leaves , leafstalks and new stalks of Paeonia lactiflora Pall. were used as explants to discuss the effect of growth regulators on inducement, multiplication , rootage and callus; and analysed the range of effective factor. Analyzing the results were as follows:1) Explants were sterilized with 0.10%Mercurie chloride (HgCl2).The result showing : The suitable time for buds was better with about 8 min. the time for petiole was better with about 12 min and for leaves was about 4 min. From November to next March was the best period of picking the explants in this test.2) The best initiation medium was 1/2MS+G A31.00mg·L-1+ 6-BA 1.00 mg·L-1 +NAA1.00mg·L-1.After 30 days culture, the inducement rate could be up to 80.00%, and G A 3 was the most important factor on inducing the caudexes.6-BA was the effective factor on multiplication, the best media was 1/2MS+6-BA1.00 mg·L-1+KT 0.50 mg·L-1.3) The effects of factors on multiplication of bud was glucose > fructose > sucrose >maltose. There are a very marked differences of glucose with maltose. 35.00 g·L-1 sucrose concentration was suitable.4) subculture time effected the multiplication and the growth of test - tube young plants. using 60 days as subculture time, theoretically, the times of propagation is 3.39. For rooting culture, medium with MS,IBA 0.50mg·L-1, IAA 0.50 mg·L-1 was the best with average length of root 1.65cm..5) For the induceing callus, leaves was the best explants. The preferable medium for the shoots callus induction was a modified MSsupplemented with 2,4D0.50 mg·L-1 and6-BA0.50 mg·L-1.
