Community structure of microbial in petroleum contaminated soil in Kelamayi Oilfield has been studied by plate colony count method, the effects of nutrition condition on the activation of stratal microflora and viscosity reduction rate were investigated by Single factor experiment and multi-factor orthogonal experimental on the based of bacteria growth and oil viscosity, comparing three DNA extraction methods, a simple operation, rapid extraction and suitable for petroleum-contaminated soil DNA template preparation have been obtained based on the DNA yield, purity, fragment size and genome integrity. Combined with Culture-free methods of molecular biology, the selected activator has been systematic evaluated. Results are summarized as follows:(1) Six types of bacteria including hydrocarbon degrading bacteria,methane-producing bacteria,nitrate reducing bacteria,sulfate reducing bacteria,sulfer bacteria and iron bacteria were isolated from five samples of oil contaminated soil in Karamay, they were able to grow in poor nutritional state and cultured 36-60h to reach the maximum growth, but their mixed bacteria growth 24h to reach the maximum ,the six types of bacteria and their mixed bacteria all have effect on oil viscosity reduction, However, the mixed bacteria have the best viscosity reducing effect, the best viscosity reduction rate is 7.90%.(2) The activator for microflora especially the beneficial bacteria and the harmful bacteria for oil recovery were Selected, it contains 0.8% sucrose, 0.45% sodium nitrate, 0.12% potassium hydrogen phosphate or potassium dihydrogen phosphate, the microbe can be effective and selective stimulated by this activator,Viscosity reduction rate of mixed bacteria could reach 46.00% on this condition.(3) Three DNA extraction methods were used for isolation of total DNA form petroleum contaminated soil. the analysis of DNA quality show that the three methods can extract corresponding DNA fragment from samples,and for the same extraction method ,different properties soil had no obvious effect on the DNA extraction; but different methods had obvious distinction in the concentration and purity of extracted DNA;Method one of the operation is simple, rapid, to extract DNA extraction rate is higher, the extracted DNA yield can achieve 98.3 ng/ul, which can be used for subsequent test.(4) The results of Culture-free molecular biology methods were as follows: the similarity of microbial community structure between samples is among the 53% -76% after activated, higher than 41%-61%, this shows that some bacteria due to lack of nutrition to growth could rapid growth under the effect of activators, and this activators could selectively activated the part of the microbial...
