Suaeda salsa is an annual herb belonging to Suaeda of Chenopodiaceae widely distributed in salinized soil, lakeside, coast, rivers and coastal regions. It is the indicator plant of saline-alkali soil due to its special morphological structure and physiological characteristics and the ability to grow in soil with high salt. Zygophyllum xanthoxylum (Zygophyllaceae) is a succulent xerophyte widely distributed in the desert regions of northwestern China with excellent adaptability to adverse arid environment. It might possess abundant genes involved in drought resistance. We have successfully cloned several genes related to the absorption, transport and compartmented, and a stable, high-efficiency plant regeneration system of Z. xanthoxylum was also established.In this study, the effects of different explants, hormone types, combinations and temperature on the callus induction of S. salsa were investigated. This work will lay the foundation for the establishment of a stable, high-efficiency plant tissue culture and regeneration system of S. salsa. And, in this study, we primarily explored the conditions of the genetic transformation and cutting propagation of Z. xanthoxylum. The work might be a good basis for further investigation of the roles of functional genes in the adaption to arid environment in Z. xanthoxylum using plant tissue culture technology combining with over-expression and RNAi technique. The main results of current study are as following: 1, Healthy callus of S. salsa were inducted, the optimal explant of S. salsa is cotyledon, the optimal hormone types are 0.1 mg·L-1 6-BA and 2.0 mg·L-1 2,4-D.2, The optimal conditions of Z. xanthoxylum transformation of the agrobacterium containing ZxNHX gene interference plasmid is pre-culture 4 d, agrobacterium infection 15 min, co-culture 2 d. The optimal conditions of Zygophyllum xanthoxylum transformation of the agrobacterium containing ZxSOSl gene interference plasmid is pre-culture 4 d, agrobacterium infection 20 min, co-culture 2 d.3, The eustipe of Zygophyllum xanthoxylum can't grow callus on the medium containing 100 mg·L-1 Carb and 70 mg·L-1 Kan, Kan can't inhibit callus tissue growth below this concentration. 4, In the cases with no hormone, cutting propagation of Z. xanthoxylum have best effect, the rooting rate is 75%.
