In this paper, Zygophyllum xanthoxylum was selected as materia. We have studied the effect of many factors on Z. xanthoxylum tissue culture, such as basic medium,light,temperature,pH, and determined the appropriate culture conditions. We have studied different explants (cotyledon,stem,eustipes,hypocotyl) and the compounding proportions of different hormones in tissue culture of Z.xanthoxylum and established a stable high-efficiency plant regeneration system.The main results were as following:(1) Compared with the B5, MS is more suitable for the basic medium of tissue culture of Z.xanthoxylum. (2) Through different explants selection, we have found that cotyledon is the best explant of callus induction and eustipes is the best explant of adventitious bud induction. (3) The best medium for callus induction was MS+0.1 mg.L-16-BA+1.0 mg.L-1 NAA, the induction rate of cotyledon was 100% and induction time was 2 weeks,2 weeks earlier than inducing by protoplasts. (4) The best medium for inducing adventitious bud was MS+1.5 mg.L-1 6-BA+0.5 mg.L-1 NAA, in which shoot differentiation rate of eustipes can reach 82.5% and induction time was 4 weeks. (5) The best medium for rooting was 1/2MS+1.0 mg.L-1 NAA+0.5 mg.L-1 IBA, in which the frequency of rooting could reach 86.7%, and in this medium, adventitious roots appeared orderly and earlier than others medium. (6) The regeneration period of Z.xanthoxylum was about 12 weeks,4 weeks less than the regeneration through proliferation.
