| Purpose：To establish chronic unpredictability burning depression merger ofacetic acid gastric ulcer in rats models, observed the expression of vanilloidreceptor subtype1(VR1) in the curative rat hypothalamus, gastric mucosa tissuemodel under electrical and the level of the rat serum gastric gastrin releasing(GAS) with the expression of5-hydroxytryptamine (5-HT), to explore themechanism of the analgesic effect of acupuncture treatment of stomach ulcersand liver stagnation utility to provide scientific theoretical basis forclinical treatment of the disease.Material and method：Select the SPF male SD rats as the research objectof this experiment, weighting140-200g, the Open-field method for behavior score,select45rats had similar, adaptive feeding for7days, were randomly dividedinto control group, the liver ulcer model group and electric acupuncture group,each group only15. In addition to the blank group, model group and electricacupuncture group with chronic unpredictable stimuli (without water for24h;break feed24h; clip tail5min; shake:1time/s,5min; day and night reversed,etc.) on depression model preparation, depression building10days combined withacetic acid burning method for the preparation of gastric ulcer model, gastriculcer after the success of the building to continue chronic unpredictabilitystimulation, stimulate the full21days. Electric acupuncture group selectedLiangqiu point and Ganshu point treatment. Electro-acupuncture treatmentinstrument given voltage2V,density wave, sparse wave4Hz, dense wave15Hz,intensity of local skin and muscle trembling slightly as the degree ofneedle, The needle was retained for20minutes, once a day, for13days. Theblank group and the model group were stimulated by sham stimulation for13days.The blank group and model group accordingly false stimulation. Observation rat generally change, open-field test results, the GUTH method to calculate thetreatment of gastric ulcer index after using Western-blot method to determinethe hypothalamus and gastric mucosa tissue content of VR1, ELISA method to detectrat gastric secrete element concentration in serum and hippocampus serotoninlevels.Results：1. Studies show that, the index of electric acupuncture group was significantlylower than that in the model group treatment outcome (P<0.01), electricacupuncture group ulcer inhibition rate of54.95%, indicating that acupuncturetherapy can promote ulcer healing of ulcers.2.Studies show that model group and blank group and gastric mucosa in rathypothalamus VR1expression significantly increased levels of statisticalsignificance (P<0.05). Electric acupuncture group improved under higher liverdepression rat hypothalamus and gastric ulcer VR1expression levels comparedwith the model group status(P<0.05).3.Studies have shown that compared with the normal group, serum gastrin detectionvalue model rats were significantly increased (P<0.01). After treatment, gastrinlevels electric acupuncture group improved compared with model group wasstatistically significant (P<0.05).4.Studies have shown that compared with the normal group, model group ratshippocampal5-HT detection value was significantly lower (P<0.01). Aftertreatment, the electric acupuncture group5-HT levels have improved, comparedwith model group was statistically significant (P<0.01).Conclusion：1.Acupuncture at Ganshu and Liangqiu points can be effectively regulated underthe liver stagnation syndrome and gastric ulcer rat hypothalamic tissuevanilloid receptor (VR1) level, improve the symptoms of liver depression ulcerpain arising.2.Effects of acupuncture by regulating the GAS and5-HT expression in hippocampus, for the treatment of gastric ulcer liver depression have a therapeutic effect. |
