Effect Of Chinese Herbal Compound On Chronic Obstructive Pulmonary Disease Complicated With Pulmonary Hypertension

Purpose：ToinvestigatetheeffectofChinese herbalcompoundvasoconstrictioninduced COPD complicated with PH in rats exposed to airway instillation of LPScombined with smoke。Material and method：1、According to the random method、50male Wister SPFrats，divided into blank control group10、model group40rats；2、Selectionof airway instillation of LPS combined exposure of rats induced by COPD withPH simulation。Method of making a model is: blank control group，model grouprats，at first14days of intratracheal instillation of model drugs instillationof0.2ml LPS (200ug/ul)， blank control group0.2ml normal saline infusion，2～13，15～70days rats in self-made animal smoked box，cigarette smoking to by30min/times (first、14days except)，2times/day，6days a week，for10weeks，the cabin with a small hole communicated with the outside。8weeks after thesuccess of the evaluation model、according to the blank control group、modelgroup、high-dose group、low dose group、western medicine group were gastriclavage、for2weeks；3、Observation and analysis of administered after the generalsituation、changes in body weight、measurement of mPAP and RVSP，aortic blood、serum after centrifugation determination of ET-1、NO and PGI2，lung tissueperfusion after taking the left lower lobe placed in fixative, to preparationof pathology；4、The data to "standard deviation"（x s）said，by single factorvariance analysis using SPSS17.0statistical software, each group two twocompared with LSD method，the P<0.05has statistical significance。Results：1.Rats in blank control group spirit in the whole process in good condition，normal，normal eating，breathing，coat gloss，color norma，physically strong， no abnormal reaction。The rats in the model group compared with the control groupappeared different degree of reduced mobility，tired of lying，four weeks afterthe rats have a cough，wheezing，shortness of breath，weight loss，hair colorand hair lose luster，feeding，drinking water is reduced，and the longer exposureto smoke，resulting in death。The treatment group rats after hair is bright，cough，breathing smoothly，activity and food intake is increased，weight gain。The effect of high dose group was improved significantly。2.Through the observation of pathological section，cytokines ET-1， NO，PGI2levels showed: the pathological findings revealed alveolar expansion，observedby HE staining and fusion：alveolar interval narrowing or fracture intofertilizer bullae。Pulmonary artery intimal hyperplasia，wall thickening，lumenstenosis，medial smooth muscle thickening，smooth muscle cell hypertrophy，exudative inflammatory perivascular associated with obvious；NO，PGI2level inthe serum decreased， increased levels of ET-1。Measured the model group ratsmPAP，RVSP significantly increased；in the treatment group compared with themodel groupET-1，NO，PGI2measured the level has improved，and mPAP，RVSP decreased。The results show that：the TCM compound prescription of rat pulmonary arteryhypertension may through protecting vascular endothelial function，improveendothelial function damage caused by dysfunction exposure on airwayinstillation of LPS combined with smoke，decrease ET-1production, increase thebody of NO，the expression of PGI2，improve the pulmonary vascular remodeling，reduce pulmonary vascular resistance and pulmonary arterial pressure and riseto protect action。Conclusion：1.Tracheal instillation of LPS combined smoke exposed rats to replicate thesuccess of COPD complicated with PH rat model.
