| Objective:To evaluate the efficacy and safety of Jingxin Zhidong Fang in treating moderate and severe tic disorders; To explore the accommodation of serum levels of IL-4, IL-5 and IL-13by Jingxin Zhidong Fang; To observe the deficiency of IL-4, IL-5 and IL-13 inmoderate and severe tic disorders; To investigate correlations between IL-4ã€IL-5 and IL-13 concentrations and symptom severity.Method:A prospective study was conducted. Patients were recruited at the Pediatric Clinic of Guang’anmen Hospital, China Academy of Chinese Medical Science.23 patients with moderate and severe TD were selected in accordance with the diagnosis and exclusion.All patients received herbal treatment of Jingxin Zhidong Fang continuously for six months. We compared the results of The Yale Global Tic Severity Scale (YGTSS) before herbal treatment versus six months after treatment. Their liver-kidney functions was monitored before and after treatment.The serum of all patients was collected at the first visit and the endpoint visit of the research. All serum samples were stored in a-80℃ freezer until assay. The assay results were compared with 14 patients without TD. Serum levels of IL-4, IL-5 and IL-13were tested by enzyme-linked immunosorbent (ELISA). Experimental data were statistically analyzed.Results:1. After 6 months treatment by Jingxin Zhidong Fang, the total effective rate was 78%. Atotal of 23 patients with moderate and severe TD, significantly improved in 11 cases,improved in 7 cases and remained in 5 cases. The total effective rate was 91% in severe TD,which was higher than moderate TD. Liver-kidney functions was all normal in 23 patientsbefore and after treatment.2. After 6 months treatment by Jingxin Zhidong Fang, serum levels of IL-4ã€IL-13 were significantly lower than before the treatment (P<0.05 both); But the difference serum levels of IL-5 before and after treatment was difficult to determined (P=0.047).3. The serum levels of IL-4ã€IL-5 was lower in patients with moderate andsevere TD than in patients without TD (P<0.05); The serum levels of IL-13 was Significantly higher in moderate and severe TD than in patients without TD (P<0.01); After 6 months treatment,No differences in the serum levels of IL-5 and IL-13 were identified between two groups(P>0.05).But the serum levels of IL-4 was Significantly lower than in patients without TD(P<0.01).4. We compared the levels of IL-4 and IL-13 in patients with moderate TD and severe TD, The levels of IL-4was negatively correlated with YGTSS. But no differences was present in the levels of IL-13. There was no statistically significant linear correlation between levels of IL-13 subclass and symptom severity.Conclusion:The clinical efficacy and safety of Jingxin Zhidong Fang in treating moderate and severe tic disorders was reliable under the condition of this study. The clinical efficacy and improvement in patients with severe TD was better than patients with moderate TD; Jingxin Zhidong Fang could reduce the serum level of IL-4ã€IL-13; The levels of IL-4 was negatively correlated with YGTSS; There were no correlations between IL-13 subclass concentrations and symptom severity. |
