After that, all the subjects were randomly divided into six groups including blank group, model group, jianpiyifei group, huatan group, jianpiyifeihuatan group and Carbocisteine group. Jianpiyifeihuatan and its separated parts were chosen as the administration drugs and the blank group was administration with pure water as well as model group. The dosage of drugs and pure water was equivalent and the time zone was 4 weeks. HE staining was used to find the morphological changes of the lung of rats by observing under optical and electron microscope. The pulmonary function tests and arterial blood gas analysis were taken to find the lung ventilation and the blood flow changes. AB-PAS and ELISA methods were separated to find the level of goblet cells and meture the content of mucins of MUC5AC and the inflammatory cytokines of IL-13〠IL-1βã€COX2ã€TNF-αã€TGF-αã€PGE2. To observe the function and difference of Jianpiyifeihuatan and its separated parts to the model rats of COPD by using the above methods.Result:Pathological observation by HE staining under optical and electron microscope:the results of observation under optical microscope and electron microscope showed that blank group had no abnormal conditions about lung tissues; compared with the blank group,the lung tissues of COPD rats had the abnormal conditions:the cilia fell off, the inflammatory cells infiltrationed and secretions increased; compared with the model group, the above pathological changes of the lung tissues had been improved including Jianpiyifei group, huatan group, jianpiyifeihuatan group, especially the Jianpiyifeihuatan group.Lung function and Aterial blood gas analysis:compared with the blank group, the relevant indicators were abnormal conditions in model group:the level of FEV0.3/FVCã€pHã€PaO2ã€SaO2 were decreased, the PaCO2 was increased; compared with the model group, the above relevant indicators had been improved including Jianpiyifei group, huatan group, jianpiyifeihuatan group, especially the Jianpiyifeihuatan group.AB-PAS and ELISA:the goblet cells had no hyperplasia and the mucins of MUC5AC were narmal as well as the relevant inflammatory cytokines of IL-13ã€IL-1β〠COX2ã€TNF-αã€TGF-αã€PGE2 in blank group; compared with the blank group, the goblet cells were hyperplasia and the content of MUC5ACã€IL-13ã€IL-1βã€COX2ã€TNF-αã€TGF-αã€PGE2 was increased. compared with the model group, the above changes had been improved including Jianpiyifei group, huatan group, jianpiyifeihuatan group, especially the Jianpiyifeihuatan group.Conclusion:In summary, Jianpiyifeihuatanfang and its separated parts could decrease the content of inflammatory cytokines, so as to suppress the goblet cells metaplasia and mucus hypersecretion, to improve the lung function and Arterial blood gas and the pathological conditions of lung tissues of model rats of COPD. but there are some differences among three Traditional Chinese Medicine group:the best is Jianpiyifeihuatan group, then group is Jianpiyifei group, last group is the huatan group. |
