DNA analysis technology becomes one of the most important basis of the criminal investigation and court trial. Accordance with the provisions of Article92of the Criminal Procedure Law of the People’s Republic of China:the summons, issue a warrant of the longest duration of not more than12hours may not be detained under the disguise of a continuous summons, issue a warrant of the suspect."And the existing DNA analysis technology takes a long time, often more than12hours. Reducing DNA testing time is now becoming a new focus of scientific research. This study was designed to establish a rapid individual recognition method for human blood by DNA-free extract, shortening the time of PCR amplification to speed up the study of the three aspects of the electrophoretic velocity in two hours to complete individual identification, rapid investigation innocent the staff of the suspects, and to minimize the harm to the innocent people.Higher rate of individual identification in this study, D3S1358, D13S317, D8S1179, D21S11, vWA, D5S818six STR loci seat and Amelogenin loci, the establishment of the rapid PCR amplification, the design and development of new rapid individual STR identification kit (Expressmarker6+1) and developed a rapid capillary electrophoresis gel EX-Q20. Collected on FTA and then take blood samples from800, using the Rapid Amplification of DNA hands-free borrowing+Expressmarker6+1+EX-Q20rapid electrophoresis analysis for DNA typing, using the Chelex100DNA extraction+Sinofiler (?) kit amplification+POP4plastic electrophoretic analysis as a parallel control, we compared the time-consuming and the results between these two technologies. The experimental results show that the PCR amplification time of the fast method is30%of the control group. During the electrophoretic analysis. fast method is also more than50percent faster than the control group. Fast time-consuming only about a quarter of the control group, a significant reduction in test time and cost for the investigation of cases and gain the initiative. At the same time, studies have shown that sub-map of the fast group is clear, the results fully consistent with the control group to prove the reliability of rapid amplification results.The results of this study reveal Expressmarker6+1rapid DNA test method was significantly faster than the conventional method, accurate and reliable results can be applied to forensic DNA identification of major cases (incident) cases of large-scale blood investigation work. |
