| [Objective] To investigate the therapeutic effects and mechanism of APC in severe acute pancreatitis-associated renal injury in rats.[Methods] Healthy adult female Sprague-Dawley (SD) rats23were randomly divided into4groups:①sham-operated group (5rats);②evere acute pancreatitis group (5rats);③treatment control group (5rats);④reated group (8rats) Sham-operated group:lightly turn the pancreas and duodenum after open,closed abdomen.; Severe acute pancreatitis group and treatment control group, treatment group:injecting5%sodium taurocholate (lml/kg) into pancreatic duct to make SAP model and18rats were screened by detecting the serum creatinine values (>98μmol/1) of the angular vein blood of rats after the injection as severe acute pancreatitis-associated renal injury in rats5hours after SAP model is made.In18rats,5rats is randomly picked as severe acute pancreatitis group; Another5rats which were injected by0.5ml physiological saline through rat tail vein is treatment control group.8rats were injected by0.5ml solution in which APC(50μg/kg) dissolve through rat tail vein as treatment group. Pancreatic and renal tissue and blood samples were gained from all animals16hours after all operation for histopathological examination and evaluation of pancreatic and renal tissue.The levels of serum amylase and creatinine and urea nitrogen were detected by biochemical analyzer. The levels of serum TNF-α and IL-8were detected by ELISA method.[Result]1Compared with treatment control group, treatment group significantly decreased the level of serum AMY (P<0.01); Compared with sham-operated group, SAP group significantly increased the level of serum AMY (P<0.01).2Compared with treatment control group, treatment group pancreas and renal injury pathology score significantly decreased (P<0.01); Compared with sham-operated group, SAP group pancreas and renal injury pathology score significantly increased (P<0.01)3Compared with treatment control group, treatment group decreased the level of IL-8,TNF-α,creatinine and urea nitrogen (P<0.01or P<0.05); Compared with sham-operated group, SAP group significantly increased the level of IL-8TNF-a,creatinine and urea nitrogen (P<0.01)[Conclusion] In the severe acute pancreatitis-associated renal injury, APC decrease the level of the IL-8and TNF-a. Through it, APC has some protective effect. |
