| Abstract:Contagious ovine pleuropneumonia is a chronic respiratory infectious disease caused by Mycoplasma ovipneumoniae,which is one of primary sheep diseases and prevalent in many countries.In recent years,respiratory disease which like contagious ovine pleuropneumonia successively broke out in several breeding Hu sheep farms in Shihezi of Xinjiang,with high incidence and death rate,and induced great economical Ioss.No such case had been reported in Xinjiang previously,therefore,it is need to ascertain pathogeny of the disease and biological characteristics of pathogen.In this study, isolation of pathogen and study on some biological characteristics was performed in several breeding sheep farms by routine methods and molecular biological methods.Three strains of Mycoplasma were isolated from seven sample of pneumonic ovine lung.It was proved that all of the three Mycoplasma isolates were Mycoplasma ovipneumoniae by morphological and cultivating characteristics,biological characteristics, serological method,and PCR method.Furthmore,this experiment mensurated the minimal inhibitory concentrations(MIC, μ g/ml) and the minimal bactericidal concentrations(MBC, μ g/ml) of ten antibacterial drugs on two Mycoplasma ovipneumoniae isolates in vitro by test tube method.The result showed that quinolones and florfenicol was most sensitive,two Mycoplasma ovipneumoniae isolates were sensitive to tylosin and lincocin but insensitive to erythromycin,the other four antibacterial drugs are hyposensitive.This study determined the existence of contagious ovine pleuropneumonia in Xinjiang and biological characteristics of pathogen isolates,exploringly established HN-PCR method for rapid detection,which can provide important science basis and technology means for fast diagnosis,molecular epidemiology,prevention and treatment of the disease.It will also has important academic value and application meaning on prevention of contagious ovine pleuropneumonia in Xinjiang. |
