Vaccinium spreads widely from tropic to the Arctic Circle. It is distributed mainly in southwest area in our country. Blueberry is used as fruit and drug that contains some useful substance to human health. Useful substance includes resisting to oxide and tan , folic acid, antibacterial agent, a great deal of fibre to eat and something else. Blueberry is fond of living in the little acid soil. Some is used as indicated plant in the acid soil. There are large areas and many kinds of acid soil in our country. The total areas amount to more than two hundred million hm2. There is one hundred million hm2 to fit for blueberry except for brick red soil and crimson soil. These areas are comparatively sterile because many crops can not adapt to acid soil, so they have great potential to be opened up.A great deal of high grade seedlings can be produced by tissue culture during a short period. It is a key factor that decides the cost is high or low to produce abundance plantlets. It is a key step to improve economic benefit by reducing cost. But surviving rate of micropropagated plantlets is low after being domesticated. It grows slowly in the field after surviving. Blueberry root system has no root hair, but its root system likes fibre. Blueberry root system has intergrowth with EM to form mycorrhiza in nature conditions. It was known that the EM played a key role in nutrient cycle in the ecosystem and promoted plant growth and also protected host plants against environmental stress.With rabbit eye blueberry being used as the main experimental materials, the experiments were carried out to study substituting TDZ for ZT because TDZ is cheaper than ZT, the problem of lower surviving rate after being domesticated and simplified medium. The physiological effect of EM to micropropagated rabbit eye blueberry plantlets were also studied by isolating EM from roots of more than ten years old Vaccinium.sprengelii Sleumer and inoculating EM to micropropagatedrabbit eye blueberry.Part one: study the factor to the initiation phase inducing rabbit eye blueberry to proliferateThe experiment results indicated: It was a suitable way that the shoots were sectioned into 20mm segments and surfaced for 6~8 min in 0.1% Hgcl2. The effect of taking material from spring was better than that from autumn.. The frequency of inducing rhizome to elongation both in autumn and in spring was higher than that of bud. The producing bud from the rhizome was stronger than that from shoot tip. Modified WPM with ZT 5.0 mg L-1 was used as inducing medium to comparatively increase inducing shoot of rabbit eye blueberry. ZT was more active than 6-BA in inducing shoot tip to proliferate. And regenerating shoot tip appeared even better to grow vigorously.Part two: study the factor to rabbit eye blueberry shoot to proliferate in the multiplication phaseIn the multiplication phase, factors that affect proliferation were studied, including hormone concentration, agar concentration and sucrose concentration, pH and subcultured duration. Experiment results showed that: modified WPM promoted rabbit eye blueberry to proliferate notably. Hormone was thought as a key factor to influence blueberry to proliferate. Modified WPM was added ZT, TDZ , GA3 and 6-BA separately. Experiment result showed that: TDZ preceded to ZT, GA3 and 6-BA, and its average coefficient of multiplication may amount to 7.0. Modified WPM added TDZ0.1mg L-1+GA31.0mg L-1+IAA0.2 mg L-1 was the best combination medium. Different sucrose concentration treatments had no notable effect on multiplication. The best medium added 20g L-1 sucrose may produce more shoots which grow vigorously later. Different sugar concentration treatments had no notable effect on multiplication. But medium was added 7 g L-1 sugar produces more regenerating shoots under pH 5.5 conditions. This result was accorded with blueberry's nature of being fond of acid soil.
