Conventional intercrossing of peach have many shortcomings,which bring biggish difficulty to breeding work. Molecular marker based on DNA overcomes these shortcomings and provides new tools for genetic research and linkage analysis.In this research , linkage map of peach was constructed with AFLP molecular marker in 109 F2 population derived from the intercross of 'Okubo' and 'Xingjin nectarine'. The main results were as follows: The stable and high efficient AFLP analysis system of peach genome was established:high quality of genome DNA was a critical step for AFLP analysis; Preamplification primer combinations of E+0/M+0 and E+0/M+1 showed no significant difference in the number of bands; the selective amplification primer combination E+2/M+3 and E+3/M+3 were both suitable to the analysis of peach genome,but the former had higher polymorphism than the latter; Silver-staining had the good virtues of low cost,short time for detecting and free of isotope contamination,and all proved that it was a safe,quick and effective detecting method.Polymorphism of parents was analysized with 128 pairs of primers,the selected 43 primers with rich polymorphism and steady bands were tested in progeny,167 markers were scored ,of which 122 markers were tallied with the map-making rule by χ2 test,the rate of effective marker was 73.05%; Using Mapmaker analytic software , 84 markers were assigned to 11 linkage groups,the rate of map-making marker were 68.85%;The whole length was 1586.1cM ,the longest group was 387.7cM,composed of 24 markers,the shortest group was 46.5 cM,composed of 3 markers;the largest average interval of groups was group 9 with interval of 26.84cM,while the smallest was group 2,7 with interval of 8.9cM;The average length of 11 linkage groups was 144.19cM,the average length between the markers was 18.88cM.
