The present study was to take inner Mongolia black glycyrrhiza as the research material.And the research had investigated the extraction of glycyrrhizic acid from glycyrrhiza,the purification teconology,the protection effect of glycyrrhizic acid against hyperlipoidemia in mice,and the adjustive effect of glycyrrhizic acid against alcohol-induced liver injury in mice.First,the research had compared four extracted methods which including hot reflux extraction,ultrasonic wave extraction,microwave extraction and ultrasonic wave microwave consociation extraction,and it optimized experimental conditions to extract glycyrrhizic acid from glycyrrhiza.It was found that the optimum extraction conditions of organic solvent extraction was as follows:power of microwave 250 watts,time of microwave 7 minutes,ratio of material and liquid 1:15.By using it,the pure degree and extraction rate respectively is up to 31.3% with 90.3%.Second,the purification technology of glycyhrrizic acid was refined bymacroreticular resins first and recrystallized by ethanol solution later was adopted.AB-8 macroreticular resin was selected from eight macroreticular resins. According to the design of single factors,the factors of effecting adsorption and elution were also researched,such as extracting solution concentration,pH,speed of adsorbent solution,effluent speed,concentration of eluting agent in order to find out the optimum conditions:the concentration of adsorbent solution 7 mg/mL, pH7,effluent speed 1 mL/min,eluent concentration of ethanol 70%.By using it,the pure degree is up to 57.6%.The glycyrrhizic acid was recrystallized by 70% ethanol solution after refined by AB-8 macroreticular resin in order to attain more pure product.The glycyhrrizic acid pure degree is up to90.2%after four recrystallization.Third,this research studied the adjustive effect of glycyrrhizic acid against hyperlipoidemia in mice.Hyperlipoidemia mouse model was set up by feed high fat feed.60 male mice were randomly divided into four groups:control group, model group,high purity glycyrrhizic acid group;crude glycyrrhizic acid group. The level of TC,TG,HDL-C,LDL-C,SOD in serum were measured.Compared with the model group,the concentration of TC,TG,LDL-C in serum of high purity glycyrrhizic acid group were decreased significantly(P<0.01;P<0.05;P<0.01) and the HDL-C,SOD were increased significantly(P<0.01;P<0.01).Glycyrrhizic acid had adjustive effect against hyperlipoidemia in mice. Finally,this research studied the protective effect of glycyrrhizic acid against alcohol-induced liver injury in mice.Alcohol-induced liver injury mouse model was set up by executive intragastric administration.60 male mice were randomly divided into four groups:control group,model group,purifier glycyrrhizic acid group;crude glycyrrhizic acid group.The level of aspartate transaminases(AST), alanine transaminases(ALT) in serum,and malondialdehyde(MDA),glutathione peroxidase(GPx) and glutathione(GSH) of liver homogenate were measured.Glycyrrhizic acid significantly reduced the elevation of AST,ALT,MDA(P<0.01;P<0.05;P<0.05) caused by alcohol,and maintained the activity of GPx and GSH(P<0.05;P<0.01).Glycyrrhizic acid had a certain degree of protective effect against alcohol-induced liver injury,and performed better than crude glycyrrhizic acid.
