| In recent twenty years, gossypol is of scientific and medical interest because of its anti-tumor, anti-fertility, and anti-viral properties. As a kind of male prophylactic, gossypol acetic acid has been applied comprehensively in many fields, such as medicine,chemical engineering and technical engineering. High-level cottonseed oil, gossypol-free cottonseed meal and methanol phase abundant in free gossypol (FG) could be obtained by the leaching process with a two-liquid phase solvent (TPS) system. If the FG in methanol phase can be extracted, prepared to gossypol acetic acid and refined to meet the medicinal criterion, the value of this technology will be greatly improved.This dissertation studied the problems in the process of extracting FG from the methanol phase after TPS treating cottonseed meat. The main issues are as follows:1,The content of free gossypol was 0.901% determined by AOCS method which was laid down by American Oil Chemistry Society . A method was developed to determine gossypol content in methanol phase from treating cottonseed meat by two-phase solvent (methanol + petroleum ether) extraction.The gossypol solved in methanol phase was resolved by transformation to acetone solvent and its supernatant liquid after centrifugation was analyzed at 370 nm. The results showed that it was a simple method with good reproducibility and its coefficient of variation was between 0.57 and 1.88. And it was a good method with high recovery rate 98.9% in determining gossypol content in methanol phase from two-phase solvent extraction.2,Cottonseed was extracted by methanol/ petroleum ether /water mix-solvents and the effects of extract time, temperature and step to extraction yield were studied.The single-step extraction experiment indicated that when the extraction temperature was 35℃, the time was 35 min and the step was two, the extraction yield was stable and was up to 85.98%.
