In this paper, a practical method for mutation breeding of Monascus strains was established, a strain with high productivity of Monacohin K was obtained, which can produce Monacohin K 41 times as much as the original strain. The characteristic of several Monascus strains, especially the strain with high productivity of Monacohin K, were studied and compared. The optimal temperature is 30-35 0C, the optimal pH is 4.0-5.5, and it can endure I 5%(V/V) ethanol in liquid medium. Method of analyzing the amount of Monacohin K by TLC and I-IPLC was studied. The results suggested that TLC can not used to determine the exact amount of Monacohin K, I-IPLC is more complex but more exact. The specification of H7PLC is: C18 column, acetonitrile-0. 1% phosphonic acid (65:3 5, V/V) used as mobile phase, the flow rate is 0.6mb/mm. The optimal technology for liquid fermentation and solid-state fermentation were studied. In liquid fermentation the best production of Monacolin K is 41.27 ii g/mL and in solid-state fermentation is 3.OSmg/g, obviously, the latter is better.
