Research On Breast Tumor Diagnosis And Pain Treatment Based On The Combination Of Tibetan And Western Medicine And Hybrid Method

Rearch Objectives: Breast tumor is a refractory disease with the highest incidence in the world.Due to its low cure rate and high mortality,the medical community has been committed to solving the problem.Up to now,many related studies have carried out,but no treatment with significant efficacy and few side effects has been discovered.There are abundant diagnosis and treatment methods of breast diseases in Tibetan medical classical literature,such as Four Medical Tantras.However,there is still no systematic research and modern basic research and clinical research.Therefore,this research is applied the theory of traditional Tibetan medicine to interpret the etiology and pathogenesis of breast cancer,and screening formula,to study the effect of alcohol extract of powder Seven Subka compound on the proliferation of human breast cancer cell MCF-7 and the activity of glucose-6-phosphate dehydrogenase(the key enzyme of pentose phosphate pathway)in breast cancer,the expression levels of G6 PD gene in normal different tissues,and normal breast and breast cancer tissues was analyzed based on GEO and TCGA data.The proliferation of MCF-7 was detected by CCK-8 kit,the G6 PD gene and protein expressions were detected by q PCR and WB experiments,the ROS content was detected by fluorescence method,the content and ratio of GSH and GSSH were detected using GSH micro assay kit.The effective treatment methods was explored,to lay a solid foundation for the research and development of anti-breast tumor and analgesic Tibetan drugs.Reseach Methods: This study adopted a mixed method of literature,experiment,and clinic.According to the theory of Tibetan medicine and clinical thinking to diagnose breast tumors,powder Seven Subka compound was selected from many therapeutic methods for breast pain relief,and its tasting-sexual-efficacy and analgesic mechanism was studied and analyzed,and the similarities between breast cancer and breast tumors in Tibetan medicine were compared.The expression level of G6 PD in normal tissues and normal breast and breast cancer tissues was analyzed by GEO and TCGA data.The proliferation of breast cancer cells was detected by CCK-8 kit.The expression of G6 PD gene and protein was detected by q PCR and WB assay.ROS content was detected by fluorescence method.The content and ratio of GSH and GSSH were determined using the kit.In the clinical study,three patients with hyperplasia and breast cancer were treated with powder Seven Subka compound for one month.The symptoms,pain index and mass size were compared before and after treatment.Research Results: 1.The Tibetan medicine belives,mammary gland size and age,menstruation,heredity,and nutrition state are concerned,mammary gland is adjacent to sternum,pectoralis major muscle,six heart arteries and veins,lymph node below clavicle the key place.The six internal cause were imbalanced by 4 inducement action and causes mammary gland tumor,belongs to spontaneity sore kind disease.According to the dialectical thinking of Tibetan medicine fever,according to the age,breast tumor belongs to the fever of adult age.According to the etiology,it is blood heat;According to the nature,it belongs to mixed heat.According to the stage,when there are no obvious symptoms at the early stage,it is hidden heat;at the middle stage,it’s increased heat;Breast ulceration overflow itching,it’s turbid heat;At late stage it is empty heat when the body is weak;According to external cause,belongs to disorder heat type.Looking from development process,breast swollen,mammary gland woman blood disease,mammary gland ruffian tumour,mammary gland tumour all belong to the four different stages of the same property disease that menstruation brings about mussily.Looking from to the treatment,for external treatment,external application of powder Seven Subka compound to soften the mass and analgesia,bloodletting to reduce blood heat,fire moxibustion to prevent diffusion,surgery to remove the mass.In terms of medicine,the decoction is used to converge and diffuse,the powder is used to reduce blood heat,epidemic heat,analgesia,and the application of muscle medicine after detrauma.For dietary therapy,to control diet and give nutritional support,avoid spicy and stimulating food.In behavior therapy,physical and mental pleasure should be maintained,lip breathing and upper limb exercise should be carried out after surgery.2.Powder Seven Subka compound taste bitter,post-taste bitter;has the effect of cold,rough,blunt,light medicine;has the function of cold,rough,blunt,light,mainly on the treatment of heat,soft,sharp,severe disease characteristics,has a curative effect on breast cancer.A total of 18 active ingredients and 227 corresponding targets were obtained by network pharmacology screening.A total of 11109 breast cancer targets were retrieved,and 211 targets were obtained after intersection of the two.17 core targets were identified by PPI network analysis.The significant items of target enrichment in biological process include response to drug,response to metal ions and response to oxidative stress,the significant items in cell components include membrane raft,microdomain and region,and the significant items in molecular function include deoxyribonucleic acid binding transcription factor binding and ubiquitin like protein ligase binding.The enriched KEGG pathways are mainly involved in lipid and atherosclerosis,PI3K-Akt signaling pathway,age-rage pathway and other pathways closely related to breast cancer.Powder Seven Subka compound can treat breast cancer through multiple components,multiple targets and multiple pathways.Among the core targets,the first one is TP53 protein,which is a nuclear localization phosphorylated protein with anti-cancer properties.P53 can inhibit the activity of G6 PD through transient interaction and induced conformation change of G6 PD.3.The expression of G6 PD gene is different between normal and breast cancer tissues,and the expression level of G6 PD gene in breast cancer tissues is higher than that in normal tissues.The overall survival rate of G6 PD with high expression was significantly lower than that of G6 PD with low expression.The ethanol extract of powder Seven Subka compound inhibited the proliferation,G6 PD activity,G6 PD gene expression,PRSS1 protein expression and GSH content of BREAST cancer Mc F-7 cells in a time-dependent and concentration-dependent manner,but increased ROS content.4.Found in clinical practice: Breast cancer case I: T4N1M1: Scab was removed on the 3rd day after external application of powder Seven Subka compound,breast skin color improved on the 8th day,approached normal color on the 25 th day,and pain was relieved.Breast cancer case II: BI-RADS IV C,external application of powder Seven Subka compound Powder for 1 month,abscess and blood overflow stopped,pain relieved,ulcer surface narrowed,and granulation tissue emerged.Breast cancer case 3: BI-RADS V,13 years after the left mastectomy,the patient applied powder Seven Subka compound externally for 1 month,and the pain disappeared in the upper and inner quadrant of the right breast,while slight pain appeared in the lower and inner quadrant.Breast swelling case 1: apply powder Seven Subka compound for 1 week,the breast pain disappeared and the texture of the lump became soft.Breast swelling case 2: fibro cyst,external application of powder Seven Subka compound for 2 weeks,the texture of the lump became soft.Breast swelling case 3: BI-RADS I: on the third day of external application,the pain was significantly relieved.After 1 month of continuous external application,the pain disappeared and the mass became significantly smaller(0.7cm*0.4cm before treatment and 0.4cm*0.32 cm after treatment).Reseach Conclusion: Through literature study,it is found in Tibetan medicine that six internal causes mammary gland swelling under the action of four external causes,and bad blood diffuses to form mammary gynecemia,gathers into mammary ruffian tumor,and then develops into intractable furuncle and carbuncle.Mammary gland tumor belongs to the category of mammary gland furuncle and carbuncle of Tibetan medicine.It should be treated in accordance with the three stages of sore diseases such as blood period,yellow water period and pus period,and the four major treatments such as external treatment,medicine,daily life and diet should be implemented.It was found that the ethanol extract of powder Seven Subka compound inhibited the proliferation of BREAST cancer Mc F-7 cells through G6 PD pathway dependent oxidative stress.G6 PD is a potential target of powder Seven Subka compound in the treatment of breast cancer.Through clinical studies,it is found that powder Seven Subka compound has a significant effect on the treatment of breast cancer pain,can heal breast cancer ulceration lesions,early breast tumors can be cured,and has a certain effect on the middle and late stage.
