The Association Study Of The Polymorphisms Of REL,TNIP1 Gene And Negative Emotion With Psoriasi

BackgroundNowdays,with the increasing of living pressure,negative emotions in the general population,such as anxiety,depression,and mental stress,are also observably increased.Long-term negative emotions may cause the disorder of body homeostasis,and subsequent physical and mental disease,which inversely increases the negative emotions,ultimately resulting in a vicious circle.Psoriasis generally needs a long treatment course and is difficult to cure,and it affects the appearance of the patients;therefore,it has a serious negative effect on the patient mental state.The pathogenesis of psoriasis is now still unclear,and the genetic factors may play important roles in its pathogenesis.Genetic studies have found lots of psoriasis susceptibility genes,including REL and TNIP1 genes,which play important roles in the pathogenesis and genetic susceptibility of psoriasis.ObjectiveTo study the correlation of the polymorphisms of REL,TNIP1 genes and negative emotion with psoriasis,and provide the important theoretical basis and guidance for the prevention and clinical treatment in patients with psoriasis.Methods419 cases of psoriasis patients and 416 healthy controls were collected from May 2014 to December 2015.Self-evaluation of the anxiety scale(SAS),self-evaluation of depression scale(SDS),and Pittsburgh sleep index(PSQI)scale were used to evaluate the negative emotions and sleep quality of subjects in the present study.The DNA was extracted from blood and the genotyping method was the ligase detection reaction for SNP rs702873 of the REL gene and SNP loci rs17728338 of the TNIP1 gene.Using epidata software for data entry,and SPSS13.0 for the statistics of descriptive data.The statistical software SPSS 13.0 and PLINK1.07 were used to analysis the genotyping data.Results:Proportions of anxiety and depression in psoriasis group were significantly higher than those in healthy control group(44.9%and 63%for cases vs.7.5%and 11.8%for controls,respectively,P = 0.000).SAS score and SDS score of psoriasis group were significantly higher than those of control group(41.33 ± 8.00 and 45.21 ± 9.06 vs.36.57± 7.66 and 29.08 ±10.36,respectively,P = 0.000).Sleep disorder proportion in psoriasis group was significantly higher than that in control group(59.2%vs.32.2%,P = 0.000).The average score of PSQI in psoriasis group was significantly higher than that in healthy control group(7.36 ± 2.31 vs.3,46 ± 2.02,P=0.000).The allele A frequency of rs702873 on the REL gene in psoriasis group was significantly lower than that in healthy controls group(9.90 vs.17.30,P<0.01,OR=0.525,95%Cl:0.393 0.701).The allele A frequency of rsl7728338 on the TNIP1 gene in psoriasis group was significantly higher than that in healthy controls group(15.16 vs.8.05,P<0.01,OR=2.039,95%Cl:1.491 2.789).The allele A frequency of rs702873 on the REL gene in psoriasis subjects with anxiety was significantly lower than that in psoriasis subjects without anxiety(6.38 vs.12.77,P<0.01).The allele A frequency of rs702873 on the REL gene in psoriasis subjects with depression was significantly lower than that in psoriasis subjects without depression(6.25 vs.15.81,P<0.01).The allele A frequency of rsl7728338 on the TNIP1 gene in psoriasis patients with anxiety was significantly higher than that in psoriasis patients without anxiety(19.68 vs.11.47,P<0.01).The allele A frequency of rsl7728338 on the TNIP1 gene in psoriasis patients with depression was significantly higher than that in psoriasis patients without depression(18.18 vs.10.32,P<0.01).ConclusionThe onset of psoriasis is closely related with negative emotions such as anxiety,depression,and sleep disorders.In the present study,the SNP rs702873 of REL gene and the SNP rs 17728338 of TNIP1 gene were confirmed to be associated with psoriasis;however they did not show any correlation with the age of onset and family history of psoriasis.The interactions between negative emotions and polymorphism locus rs702873 of REL gene and polymorphism locus rsl7728338 of TNIP1 gene were closely associated with psoriasis,suggesting that negative emotions may play a vital role in the occurrence,development and recurrence of psoriasis.
