Objective:Traditional Chinese Medicine focuses on syndrome differentiation and treatment.The research here proceeds with the protective effects and the mechanism on myocardial ischemia reperfusion injury in rats based on the combinaton of the hypericum attenuatum choisy and angelica,starting with Traditional Chinese Medicine theories and experiments,It aims at improving and controlling the development of the disease in order to promote the life-quality of the human being.Methods:1.Combinate the hypericum attenuatum choisy and haplo-angelica according to 1:1,1:2,2:1 on myocardial ischemia reperfusion injury rats by these matches to observe the CK content,the active effects of LDH and myocardial pathology of model rats by light microscope,screening out the combination with the best protective effects on MIRI rats.2.Observe the GSH-Px,Ca2+Mg2+-ATP,Na+K+-ATP enzyme activity in MIRI rats and the expression of Bcl-2,Bax,Hsp70 and NF-κB65,and mRNA,exploring the mechanism against myocardial ischemia-reperfusion injury through the combinaton of the hypericum attenuatum choisy and angelica.Results:1.The hypericum attenuatum choisy and angelica ratio at 2:1 can obviously decrease the serum levels of CK and the LDH activity in rats.It proves to be the combination with the best protective effects against MIRI rats improving myocardial pathological morphology and decreasing the cardiomyocyte apoptosis in rats.2.The hypericum attenuatum choisy and angelica ratio at 2:1 can obviously promote the vitality of GSH-Px in myocardium and Ca2+Mg2+ATP,Na+K+-ATP in MIRI rats,raising Bcl-2 protein expression and lowering expression of Bax.3.The hypericum attenuatum choisy and angelica ratio at 2:1 can obviously increase the exression of HSP70,Cx43,diminish the activity of NF-κB p65,enhancing the expression of HSP70 mRNA,Cx43mRNA and inhibiting NF-κB p65 mRNA.Conclusions:1.The hypericum attenuatum choisy and angelica ratio at 2:1 can obviously decrease the serum levels of CK and the LDH activity in rats.It proves to be the best combination ratio against myocardial ischemia reperfusion injury in rats improving myocardial pathological morphology and protecting the myocardium of rats.2.The hypericum attenuatum choisy and angelica ratio at 2:1 can obviously promote the vitality of GSH-Px in rats myocardium and Ca2+Mg2+-ATP,Na+K+-ATP indicating that its functional mechnism and the degree of freeing oxy radicals and enhancing the anti oxidation are associated with the lighteningof calcium overload.The protectional dynamic is reached to the myocardial ischemia reperfusion injury in rats.3.The hypericum attenuatum choisy and angelica ratio at 2:1 can obviously raise the protein expression of Bcl-2in model rats and lower Bax presenting its role of suppressig the myocardial ischemia reperfusion injury by conrolling cells apoptosis.4.The hypericum attenuatum choisy and angelica ratio at 2:1 can evidently increase the expression of HSP70,Cx43 and mRNA,lessing the protein activation of NF-κB p65.It indicates that the function against MIRI is associated with the promotion of HSP70,the restraing of NF-κB channel and cardiomyocyte apoptosis or the damage of lipid peroxidation.
