Effect Of Bushen Huoxue Formula On IL-1β、DDR2 And MMP-1、MMP-7 Expression In Rat Osteoarthritis Models And Observation On The Clinic Effect In Osteoarthritis

Objects:1.To investigate the effect of Bushen Huoxue Formula on the apoptosis of articular chondrocytes in a rat model of osteoarthritis by observing the changes of articular cartilage and the level of IL-1β in serum and joint synovial fluid,the level of DDR2 in articular cartilage cells,the expression level of MMP-1mRNA,MMP-7mRNA in joint synovial cells after ostcoarthritis model rats were treated with Bushen Huoxue Formula.2.To explore the clinical advantage of traditional Chinese medicine preparation in treating knee osteoarthritis by observing the clinical effect of Bushen Huoxue Formula on osteoarthritis of knee joint.Methods:1.Experimental study:SD rats were used to establish the osteoarthritis model by classical surgery with Hulth’s method before the gas-tric gavage treatments.Model rats were divided into Bushen Huoxue Formula treated,Glucosamine sulfate capsule treated and non-treated control groups and were gastric gavaged with Bushen Huoxue Formula,Glucosamine sulfate capsule and Starch suspension for 6 weeks respectively.To observe the general situation of rat articular cartilage by naked eye and the microscope,and the level of IL-1β,the level of DDR2,the expression level of MMP-ImRNA and MMP-7mRNA which were detected by ELISA,ABC,RT-PCR respectively.2.Clinical study:Patients with osteoarthritis of the knee were randomly divided into two groups:the recipients of Bushen Huoxue Formula and the Glucosamine sulfate capsule.After 6 weeks of treatment,the patients’ treatment effects were scored by the Improved Lequesne osteoarthritis severity evaluation index and the joint synovial fluid levels of TNF-α,IL-β and MMP-1 were measured before and after the treatment.Results:1.Experimental study:Chinese medicine,Western medicine treatment groups of rat articular cartilage observation and microscopic observation results were significantly better than the non-treated control group.The level of IL-1β in serum and joint synovial fluid,the level of DDR2 in articular cartilage cells,the expression level of MMP-1mRNA,MMP-7mRNA in joint synovial cells of the rats in the experimental group were compared as follows:The level of IL-1β in serum and joint synovial fluid,the level of DDR2 in articular cartilage cells,the expression level of MMP-1mRNA,MMP-7mRNA in joint synovial cells in the treatment groups were significantly higher than those in the blank control group(P<0.05),there was no significant difference in the level of IL-1β,the expression level of MMP-1mRNA and MMP-7mRNA between the two treatment groups(P>0.05),but there was significant difference between the two treatment groups in the level of DDR2(P<0.05).2.Clinical study:There was no significant difference in the score of Improved Lequesne osteoarthritis severity evaluation index between Bushen Huoxue Formula group and glucosamine sulfate capsule group(P>0.05).There was no significant difference between the two groups before and after treatment in the levels of TNF-α,IL-1β(P>0.05),but there was significant difference between the two groups in the level of MMP-1(P<0.05).Conclusions:1.Bushen Huoxue Formula can inhibit the secretion of IL-1β in synovial cells and expression of osteoarthritis signal factor DDR2,meanwhile reduce the gene expression of MMP-1mRNA and MMP-7mRNA in synovial tissue.Experimental study confirmed that Bushen Huoxue Formula on osteoarthritis rat model has a clear therapeutic effect,which may be the mechanism of Bushen Huoxue Formula treatment on osteoarthritis.2.The Improved Lequesne osteoarthritis severity evaluation index of patients with osteoarthritis of the knee was significantly improved after taking Bushen Huoxue Formula,and the levels of TNF-α,IL-β and MMP-1 in joint synovial fluid of patients with Bushen Huoxue Formula significantly reduced,which showed a clear knee osteoarthritis clinical treatment.
