Objective:Used a model of middle cerebral artery occlusion(MCAO)of the rats,intervention by electroacpuncture,acupuncture point were selected the Baihui and Shenting.And then the blood flow?the blood volume?the mean transit time and glucose metabolism of the rats were detected by micro Magnetic Resonance Imaging and micro positron emission tomography.Then the leaning and memory function of the rats were detected,and study its possible mechanisms of the electroacpuncture protective effect.Method:80 male Sprague-Dawley rats were randomly divided into sham operation group(20 rats)and MCAO model group(60 rats).Model of focal cerebral ischemia injury of the rats were used suture method to make,then the rats were randomly divided into model group,electroacpuncture group and non acupoint group,which were 20 in every group.Electroacupuncture group and non acupoint group rats began to intervene in 24 hours after the operation.Electroacupuncture points included Baihui and Shenting,non acupoint group take bilateral threat.The EA group using density wave,frequency 1-20hz,30 minutes each time,1 times/day,sustain intervention 7 days.The rats of the sham operation group and model group with the same grasping and fixed 30min,would not be stimulated.After 24 hours,3 days,5 days,7 days,the rats were respectively scale of marks by Zea longa,then the neural function defect of the rats were observed.To observe the ability of the learning and memory of the rats,which were detected by Morris water maze.After the 7th day,the volume of cerebral infarction of the rats were detected by small animal magnetic resonance.And also the cerebral blood flow(CBF)?cerebral volume(CBV)and mean transit time(MTT)were detected by the perfusion weight imagic(PWI)series of micro MRI,before the detected the rats which were injected the"DIMEGLUMINE GADOPENTETATE INJECTION(Gadopt)"in the tail vein of the rats.The CBF?CBV and MTT which were reflected the effects of electroacpuncture Shenting and Baihui,around the ischemia core zone,peripheral zone and damage zone of the rats.And the mean time,after the 7th day of the EA or intervene the rats,then detected the brain glucose by micro PET,which injected 18F-FDG in the tail vein.Results:1.The score of neurological behavior:The neurobehavioral score of the electroacupuncture group compared with the model group,non acupoint group,the difference was obvious,which indicating that EA Baihui,Shenting acupoint can improve the symptoms of neurological deficits in the MCAO rats.2.Water maze experiment:Directional navigation experiments:The escape latent of the EA group was less than those rats in the model group and non acupoint group,the difference was obvious.Space exploration experiments showed that,acrossing the platform of which the rats in the EA group were more than those in the model group and non acupoint group,the difference was obvious.The results indicated that the learning and memory ability of the MCAO rats could been improved by EA Baihui,Shenting acupoint3.The volume of cerebral infarction:The cerebral infarction volume was detected by using T2 series of the small animal magnetic resonance,the cerebral infarction volume in EA group was less than the model group and non acupoint group,the difference was obvious.The results indicated that the cerebral infarction volume of the MCAO rats could been reduced by EA Baihui,Shenting acupoint.4.Cerebral blood changed:CBF value:Cerebral blood flow in the EA group was faster than the blood in model group and non acupoint,the difference was obvious.CBV values:Cerebral blood volume in the EA group was more than the blood in model group and non acupoint,the difference was obvious.MTT:The MTT in the EA group was faster than it in model group and non acupoint,the difference was obvious.The results indicated that the regional cerebral blood flow(CBF),blood volume(CBV),mean through time(determined by MTT)could been increased by EA Baihui,Shenting acupoint5.Glucose metabolism:Micro PET-CT showed,the glucose metabolism in EA group was significantly higher than that of in the model group and non acupoint group rats,the difference was obvious.The amygdala,caudate putamen,anterior cingulate cortex,entorhinal cortex,anterior insula,medial prefrontal,orbitofrontal cortex,posterior cingulate gyrus,anterior dorsal hippocampus,which were the cognitive related brain regions,the glucose metabolism of the brain regions were changed.The results indicated that the glucose metabolism of the MCAO rats could been improved by EA Baihui,Shenting acupoint.Conclusion:1.The ability of learning and memory of the MCAO rats could been improved by electroacupuncture Baihui,Shenting acupoint.2.The CBF?CBV?MTT of the MCAO rats could been improved by electroacupuncture Baihui,Shenting acupoint.3.The glucose metabolism of the MCAO rats could been improved by electroacupuncture Baihui,Shenting acupoint.4.The glucose metabolism of the rats,included.the amygdala,the caudate putamen,cingulate,entorhinal cortex,insula before,the medial prefrontal cortex,medial orbitofrontal cortex,dorsal hippocampus before,dorsal hippocampus and other related cognitive area could been improved by electroacupuncture Baihui,Shenting acupoint.
