Effect Of Electroacupuncture On Synaptic Plasticity By JAK2-STAT3 Signaling Pathway On Spatial Learning And Memory In MCAO/R Rats

Objective:The object of the study is to explore the mechanism of electroacupuncture on DU20 and DU24 acupoints on the improvement of spatial learning and memory of rats with MCAO/R possibly by regulating JAK2-STAT3 pathway which can influence synaptic plasticity.Methods:80 SD male rats with SPF level were used in the study among which 62 rats were performed MCAO/R by thread embolism method and another 18 rats were only performed vascular separation as a sham group.24 hours after model establishment,rats were undergone MRI and jumping stand experiment to calculatecerebral infarct volume and evaluate the cognitive disorder of rats respectively.Finally 54 rats with MCAO/R and cognitive disorder were randomly divided into 3 groups including the model group(n=18),the electro-acupuncture group(n= 18)and the non-acupoint group(n=18).After model establishment,After model establishment,rats in the electroacupuncture group were performed electroacupuncture intervention on DU20 and DU24 acupoint and and non-acupoint group was electroacupuncture at non-acupoint for 30 minutes with 2/20HZ and 1-3mA for 14 days.Zea-longa score was used to observe neurobehavior of rats before intervention,1st day,7th day and 14th day post-intervention.Morris water maze was used to record the spatial learning and memory of rats from 10th day to 14th day post-intervention.Jumping stand experiment was used to evaluate the memory of rats on 14th day post-intervention.Finally,MRI was used to calculate the volume of cerebral infarction of rats.After sacrifice,PSD-95 and SYN expression were detected by immunohistochemical.Ultrastructure of synaptic on the left hippocampus district was detected by transmission electron microscope.After sacrifice,PSD-95 and SYN expression were detected by immunohistochemical.Ultrastructure of synaptic on the left hippocampus district was detected by transmission electron microscope.The expression and phosphorylation of JAK2 and STAT3 by western blot.Finally,18 rats in each group were included for statistic.Results:(1)MCAO/R:The volume of infraction:there is no cerebral infraction detected by T2W1 in sham group while cerebral infraction was detected in another three groups with no distinction(P>0.05).On the 14th day after intervention T2W1 can shows that the volume of cerebral infraction in electroacupuncture is smaller than that of model group and non-acupoint with significant statistic significance(P<0.05).(2)Zea-longa neuroethology score:On the 7th day and 14h day after intervention,Zea-longa score of electroacupuncture goup was better than that of model group(P<0.01)and non-acupoint group(P<0.05).Jumping stand experiment:there is no distinction of the standing time of rats in electroacupuncture group,model group and non-acupoint group(P>0.05).While,on the 14th day after intervention,compared with sham group and non-acupoint group,standing time of rats in electroacupuncture group is longer with significant statistic significance(P<0.05).Water morris test:The escape latency time in the electroacupuncture group is shorter than model group and non-acupoint group with significant statistic group(P<0.05).Through the platform number in sham group is more than that of another three groups with significant statistic significance(P<0.05)(3)Immunohistochemistry:PSD-95 and SYN expression in the left hippocampus CA1 district:compared with sham group and non-acupoint group,model group is lower with significant statistic significance(P<0.05);compared with model group and non-acupoint group,PSD-95 and SYN expression in the electroacupuncture group is higher with significant statistic significance(P<0.05).(4)Transmission electron microscope:compared with sham group and non-acupoint group,model group is lower with significant statistic significance(P<0.05);compared with model group and non-acupoint group,the synaptic number of electroacupuncture group is more with significant statistic significance(P<0.05)in the left hippocampus CA1 district.(5)Western blot:In the left hippocampus district,compared with sham group and non-acupoint group,model group is higher with significant statistic significance(P<0.05);compared with model group and non-acupoint group,p-JAK2 and p-STAT3 expression in electroacupuncture group is lower with significant statistic significance(P<0.05).Conclusion:Electroacupuncture at DU20 and DU24 increased the expression of PSD-95 and SYN and other important synapse related protein,promoted the integrity of the synaptic ultrastructure and improved synaptic plasticity may by inhibiting the activation of JAK2-STAT3 pathway.
