[Purpose] The current treatments for Parkinson’s disease (Parkinson’sdisease, PD) continue to be limited. As an experienced clinician, ProfessorMinghui Yang has unique insights into treatment of PD with methods ofstrengthening kidney and activating blood circulation. We performed the study tosummarize the academic experience of Professor Yang on PD’s treatments,explored the mechanism of strengthening kidney and activating blood circulationmethods in order to provide the objective basis for treating PD with the traditionalChinese medicine.[Methods] We comprehensively summarized the academic experience ofProfessor Yang on PD’s treatments from theory to practice by attending courses,following the outpatients, collecting and organizing medical records and relateddocuments. The120SD rats were randomly divided into the model group (100rats) and the normal control group (20rats). The100rats were modeled by6-OHDA methods and54rats were successfully modeled. The54modeled ratswere randomly divided into the treatment group (30rats) and the control group(24rats). The treatment group was intragastrically administrated withBushenhuoxue granules for eight weeks, while the control group withphysiological saline for eight weeks. We observed the apomorphine(APO)-induced behavior changes before and after treatment. The expressions anddistributions of adenosine A2Areceptor, DMTI, VMAT2, Nrf2protein wereexamined by immunofluorescence and immunohistochemistry in the striatum andsubstantia nigra of rats.[Results] Professor Yang developed the traditional Chinese medicine theorythat wind syndrome with tremors and vertigo was mainly due to the liver disorder.He proposed “deficiency of kidney and blood stasis” and believed that was themain pathogenesis of PD. The therapeutic choice of strengthening kidney andactivating blood circulation method showed evidence of clinical benefits.Strengthening kidney and activating blood circulation granules significantly improved the rotational behavior of PD rats. Compared with the control group, thenumber of rotations was significantly reduced in the treatment group (p <0.01).Immunofluorescence showed the expression of adenosine A2Aand DMT1proteinin striatum area was increased compared with the normal control group, wasdecreased relative to the model control group. The expression of Nrf2andVMAT2protein of treatment group was reduced compared with the normalcontrol group and was increased relative to the model control group.Immunohistochemistry showed that adenosine A2Aprotein was not expressed insubstantia nigra region of the rat brain. The expression of DMT1protein insubstantia nigra region was increased compared with the normal control groupand was decreased relative to the model control group. The expression of Nrf2and VMAT2protein of treatment group in substantia nigra region was reducedcompared with the normal control group and was increased relative to the modelcontrol group.[Conclusions] Professor Minghui Yang has formed his own theoreticalsystem and treatment philosophy of PD. The therapeutic choice of strengtheningkidney and activating blood circulation method showed evidence of clinicalbenefits. Bushenhuoxue granules significantly improved the rotational behavior ofPD rats by decreasing the expression of adenosine A2Aand DMT1protein andincreasing the expression of Nrf2and VMAT2protein. The Bushenhuoxuegranules also enhanced the activity of dopamine D2receptors and spiny neurons,decreased the abnormal deposition of iron in the brain, regulated the number ofDA in the synapse and reduced the toxic damage to the dopaminergic neurons toexert the antioxidant and antagonistic function to A2Areceptor. The mechanismsinvolved above cooperated to make the therapeutic effect of Bushenhuoxuegranules on PD.
