PrefaceThe invasion and metastasis of tumor is the most important and essential biological characteristic of malignant tumor,which is mail reason to induce unfavourable prognosis and death.The incidence and the mortality of gastric cancer is still very high in our country and even all the world.There is no specific symptom in early gastric cancer.The most patients of gastric cancer were in the advanced stage when they visited doctors,which often dead of extensive invision and metastasis. Therefore,the researth of immunity molecule and genes correlated with the invision and metastasis of gastric cancer can provide assistants and evidents to early diagnosis and targeted therapy of the invision and metastasis of gastric cancer.The invision and metastasis of gastric cancer is a multi-procedures complex event in essence,it includes, l,the change of adhension within tumor cells and normal cells,extracellular matrix(ECM).2,the increase of extracellular matrix degradation.3,the migration of tumor cells.4 the neovascularization.5,the proliferate of tumor cells which escaped from the host immunologic surveillance.But now the mechanism of the invision and metastasis of gastric cancer is not yet identify.It is very important for the early diagnosis,treatment instruction and the evaluation of prognosis to searth and study the changes of express and the functions of genes and pruductions correlated witn the invision and metastasis of gastric cancer.We study the express of heparinase,matrix metalloproteinase MMP-2,MMP-14 and cadherin in the gastric cancer tissue and perigastric normal mucosal tissue,and make a statistics analysis between heparinase, matrix metalloproteinase MMP-2,MMP-14,E-cadherin and gastric cancer biological behavior.We hope to knom the possibility that heparinase,matrix metalloproteinase MMP-2,MMP-14,E-cadherin are regared as clinical screen index of the gastric cancer recrudescence or metastasis.Methods1.The collection of gastric cancer operational sample and pathological paraffin block in our hospital from Apr 2007 to Aug 2007.Together with the cancer tissues in 67 patients,adjacent normal gastrie tissues in 67 patients were resected,Each sample was Snap-frozen and stored at-80℃.2.The express of heparinase,matrix metalloproteinase MMP-2,MMP-14, E-cadherin in the gastric cancer tissue and perigastric normal mucosal tissue were detected in immunohistochemistry.3.The express of heparinase,matrix metalloproteinase MMP-2,MMP-14, E-cadherin in the gastric cancer tissue and perigastric normal mucosal tissue were detected in Western blot.4.The express of heparinase,matrix metalloproteinase MMP-2,MMP-14, E-cadherin in the gastric cancer tissue and perigastric normal mucosal tissue were detected in RT-PCR.5.Statistics data were analyzed by X~2-test,t-test and spearman methods of SPSS 10.0 software.Results1.Heparinase shows high expression in gastric cancer tissues.The results of immunohistochemistry show that the express of heparinase is 76.1%in gastric cancer tissues and 20.9%in gastric normal tissues.The positive rate in cancer tissues was significantly higher than that in normal tissues(P<0.01).The results of Western blot show that the express of heparinase is 77.6%in gastric cancer tissues and 23.9%in gastric normal tissues(P<0.05).The results of RT-PCR show that the express of heparinase mRNA is 74.6%in gastric cancer tissues and 31.3%in gastric normal tissues(P<0.05).It is high expression in cancer tissues showed a significantly differentiation in gross appearanee,depth of invasion,cell differentiation level, histology and growth stale,lymph node metastasis and TNM stage,but no significant differences in tumor size.2.The results of immunohistochemistry show that the express of MMP-2 is 85.1%in gastric cancer tissues and no express in gastric normal tissues.The positive rate in cancer tissues was significantly higher than that in normal tissues(P<0.05).The results of Western blot show that the express of MMP-2 is 88.1%in gastric cancer tissues and 16.4%in gastric normal tissues(P<0.05).The results of RT-PCR show that the express of MMP-2 mRNA is 83.6%in gastric cancer tissues and 56.7%in gastric normal tissues(P<0.05).It is high expression in cancer tissues showed a significantly differentiation in depth of invasion,cell differentiation level,lymph node metastasis and TNM stage,but no significant differences in tumor size,gross appearanee,and histology and growth stale.3.The results of immunohistochemistry show that the express of MMP-14 is 91.0%in gastric cancer tissues and no express in gastric normal tissues.The positive rate in cancer tissues was significantly higher than that in normal tissues(P<0.05).The results of Western blot show that the express of MMP-14 is 94%in gastric cancer tissues and 19.4%in gastric normal tissues(P<0.05).The results of RT-PCR show that the express of MMP-2 mRNA is 77.6%in gastric cancer tissues and 43.3%in gastric normal tissues(P<0.05).It is high expression in cancer tissues showed a significantly differentiation in depth of invasion,cell differentiation level,lymph node metastasis and TNM stage,but no significant differences in tumor size,gross appearanee,and histology and growth stale.4.The results of immunohistochemistry show that the express of E-cadherin is 55.2%in gastric cancer tissues and 94%in gastric normal tissues.The positive rate in cancer tissues was significantly higher than that in normal tissues(P<0.05).The results of western blot show that the express of E-cadherin is 50.7%in gastric cancer tissues and 97%in gastric normal tissues(P<0.05).The results of RT-PCR show that the express of E-cadherin mRNA is 86.8%in gastric cancer tissues and 98.5%in gastric normal tissues(P<0.05).It is low expression in cancer tissues showed a significantly differentiation in depth of invasion,cell differentiation level,lymph node metastasis and TNM stage,but no significant differences in tumor size,gross appearanee,and histology and growth stale.5.There is relativity between the high expression of MMP-2,MMP-14 in gastric cancer tissues,but no relativity within the expression of MMP-2,heparinase and cadherin.Conclusion1.Heparinase shows high expression in gastric cancer tissues and is related to gastric cancer biological behavior and prognosis judgement.2.MMP-2 shows high expression in gastric cancer tissues and is related to gastric cancer biological behavior and prognosis judgement.3.MMP-14 shows high expression in gastric cancer tissues and is related to gastric cancer biological behavior and prognosis judgement.4.E-cadherin shows low expression in gastric cancer tissues and is related to gastric cancer biological behavior and prognosis judgement.5.The united detection of the heparinase,matrix metalloproteinase MMP-2, MMP-14,E-cadherin will be helpful to judge prognosis and formulate treatment program.
