DNA Ploidy Analysis And The Expression Of Matrix Metalloproteinase-9, Tissue Inhibitors Of Matrix Metalloproteinase-2 And Epithelial-cadherin In Gastric Cancer

Objective This study aimed to investigate DNA ploidy and the expression of MMP-9 ,TIMP-2 and E-cadherin in gastric carcinoma,for the view of neoplasm heterogeneity ,to explore the machinery of molecular biology and genetics for gastric carcinoma.Methods 1 Immunohistochemical SP methods : Using monoclonal antibodies for MMP-9 ,TIMP-2 and E-cadherin ,we examined 99 cases of gastric carcinoma,16 cases of benign gastric tissue around gastric carcinoma, 16 cases of distant metastasis tissue and 25 cases of metastastic lymph node from gastric. The results is treated by statistics methods -chi-square test .2 Flow cytometry : Flow cytometry DNA ploidy and S-phase fraction (SPF) analysis was performed on 47 cases of gastric cancer ,6 cases of benign gastric tissue around gastric carcinoma and 4 cases of distant metastasis tissue with the use of paraffin-embeded sections .Results There is a negative relationship of MMP-9 expression with deferenatiation degree, and tumor formation ,but a positive relationship with the depth of invasiveness(in mucous membrane, infra- mucous membrane to muscular layer and adventitia, MMP-9's positive rate is 28.6%,59.3%,and 73.8% respectively,P<0.05) ,lymph node metastasis(in lymph node metastasis negativation and positivation, MMP-9's positive rate is 54.3% and 79.2% respectively,P<0.01), tumor and nontumor(in gastric cancer and mucous membrane around gastric cancer,MMP-9's positive rate is 66.7% and 31.3%respectively, P<0.01) , tumor and metastastic lymph node and TNM stage .There is a negative relationship of TIMP-2 expression with deferenatiation degree , tumor formation, tumor and nontumor and metastastic lymph node,but a positive relationship with the depth of invasiveness ( in mucous membrane, infra-mucous membrane to muscular layer and adventitia, TIMP-2's positive rate is 71.4%, 59.3% and 33.8% respectively,P<0.05 ) ,lymph node metastasis(in lymph node metastasis negativation and positivation, TIMP-2's positive rate is 56.5% and 32.1% respectively,P<0.05) .There is a negative relationship of E-cadherin expression with TNM stage and tumor formation, but a positive relationship with the depth of invasiveness(in mucous membrane, infra- mucous membrane to muscular layer and adventitia, E-cadherin's positive rate is 85.7%. 48.1% and 35.4% respectively, P<0.05 ),lymph node metastasis(in lymph node metastasis negativation and positivation, E-cadherin's positive rate is 54.3% and 32.1% respectively, P<0.05), tumor and nontumor(in gastric cancer and mucous membrane around gastric cancer, E-cadherin's positive rate is 42.4% and 87.5%, P<0.001), tumor and metastastic lymph node and deferenatiation degree.There is a positive relationship between MMP-9 and TIMP-2 and between MMP-9 and E-cadherin,but a negative relationship between TIMP-2 and E-cadherin.There is a positive relationship of flow cytometry DNA ploidy and SPF with the deferenatiation degree lymph node metastasis and tumor and nontumor, but a negative relationship with the depth of invasiveness , tumor formation and TNM stage.The relationship between immunohistochemistry and flow cytometry: There is a negative relationship of DNA ploidy and SPF with MMP-9 andTIMP-2, but a positive relationship with E-cadherin.Conclusion: It is a complex program for tumor progression and heterogeneity ,and MMP-9 ,TIMP-2 , E-cadherin, DNA ploidy and SPF participate in the program together.
