| Objective:To investigate the effects of brucea oil on the treatment of psoriasis and the influence of Th1/Th2 balance and keratinocytes proliferation.Methods:Study on the influence of Bruceae oral emulsion on animal model of Thl disease. First we construct animal models of Thl-type disease that is autoimmune thyroiditis (EAT), which were randomly divided into groups EAT model,triptolide treatment group, Brucea javanica oil emulsion oral treatment group. Then, the rat were killed and the lever of TgAb,IL-18 and IL-10 in serum were detected respectively, pathological changes of thyroid were observed with light microscope. Stdudy of Bruceae oral emulsion on the mitosis of mice's basal cells in the vaginal epithelium and the formation of mouse-tail epidermis granular layer. Kunming mice were divided into three groups, the normal saline treatement as negative control group (NS), methotrexate treatment as positive control group (MTX) and Bruceae oral emulsion group as the treatment group. Rats were executed and pathology pictures were made.The mitotic index of basal cells and the Scales with granular layer in per hundred Scales were calculated, and then inter-group comparsons were used. Study on Bruceae oral emulsion on psoriasis-like pathological changes of guinea pigs'ears. There were five groups, the noemal control group, model group, Bruceae oral emulsion group, and saline treated as negative control group (NS), methotrexate treated as positive control group (MTX).Rats were executed and then pathology pictures were made and detected to compared baker grade and cellular infiltrates and to explord pathomorphism changes of heart, liver, lungand kidney in experimental animals. Study on the effects of Bruceae oral emulsion on psoriasis therapy and Th1/Th2 balance in serum. Psoriasis vulgaris were divided into experimental group and control group, normal group, experimental group were given oral Brucea javanica oil emulsion, the control group received Antaisu compound for 8 weeks. Efficiency and efficacy index were calculated respectively 8 weeks later on the basis of PASI grade reduction rate and inter-group comparsons were used. The serum lever of IL-18 and IL-10 in each group should be detected at the same time and the changes and the ratio should be recorded. Blood routine, urine routine, stool routine, liver function, kidney function and so on should be checked after therapy.Results:Study on Brucea javanica oil's efficacy in Thl-type animal model: Compared with untreated EAT model group, the TGAb level and IL-18 of treated groups of the rat model of EAT treated with tripterygium polyglucoside and Brucea javanica oil decreased and IL-10 increased remarkably (P<0.01),there is no difference between the rat model of EAT treated with tripterygium polyglucoside and Brucea javanica oil (P> 0.05). But as for the improvement of EAT,there was no statistic significance between EAT group and each treated group (P>0.05). Study on the effect of Brucea javanica oil on mitosis of mouse vaginal epithelium and formation of granular layers in mouse tail-scale epidermis:Compared with MTX group, Brucea javanica oil treated group and NS group, the mitosis index and the number of squama with granular layers in every 100 squama, had significance difference, but MTX group and Brucea javanica oil treated group had no significant difference (P>0.05).Study of Brucea javanica oil on Psoriasis-like pathological of guinea pig. The Baker scores and the number of inflammatory infiltration cells of Brucea javanica oil treated group decreased to different level, which was significantly different with that of NS group and model control group, but compared with MTX group, there were no significant difference.Study on the efficacy of Brucea javanica oil on psoriasis and the influence to the balance of Thl/Th2 in peripheral blood. Serum IL-18 and IL-10 levels and their ratios were significantly different between Patients and normal control group.The efficiency of patients treated with Brucea javanica oil for 8 weeks was 80% and efficacy index was 70.14±22.89, which suggested the effect of Brucea javanica oil and compound ampeptidum had no significant difference.serum IL-18 level declined after treatment, showing significant difference compared with before-treatment, IL-10 level rised, showing no significant difference compared with before-treatment, and the ration of IL-18 and IL-10 declined, showing significant difference compared with before-treatment. Blood routine test, urine routine test, stool routine test, and the liver and renal functions in patients after treatment were normal.Conclusion:The actions of Brucea javanica oil is safe and active against psoriasis. It can correct the Th1/Th2 shift of patients with psoriasis, and inhibit the keratinocyte cell hyperplasia. It is suggested that correcting the parakeratosis is the main mechanism. |
