Methods To treat rats with glucocorticoid and vitamin after having duplicated animal model of allergic rhinitis; to compare sneeze quantity and serum level of four cytokines and IgE by enzyme linked immunosorbent assay (ELISA) with normal control; to detect the expression of recombination activating gene-1 (RAGs) and DNA sequence of RAGl in nasal membrane and thymus which are from AR group, DEX group and normal control by reverse transcription polymerase chain reaction (RT-PCR) and DNA analysator. Results The sneeze quantity in AR group is notablely higher than those in normal control, vitamin E group, vitamin AD group and DEX group (P<0.05); the serum level of IL-4, IL-10 and IL-17 in AR group is significantly difference from those in normal control, vitamin E group and DEX group(P<0.05); the serum level of INF-γ and IgE in AR group is significantly difference from those in normal control, vitamin E group and DEX group(P<0.05) except in vitamin AD group(P>0.05); there are no RAGl mRNA expression in normal nasal membrane in contrast with what happens in AR the expression is higher than in DEX group; the expression of RAGl is all positive in the thymus of three groups; the homology of DNA sequence is 100% between determine sequence and known sequence. Conclusion These results confirm that there is dominant differentiation of Th2 cell in allergic rhinitis and RAGl is related with allergic rhinitis which can accommodate the secretion of four cytokines through influencing the differentiation of T help cell but there are not mutation and deletion on the process of allergic rhinitis. VitE can drop down the level of IgE through changing the secretion of IL-4 and INF-γ but VitAD has no this power. Glucocorticoid is an effectively immunomodulator and good for relieving the symptom of allergic rhinitis through several ways but cannot change the expression of RAGl.
