Objective: To study the effect of acupuncture therapy of " YiQffiaoXue ,FuBenPeiYuan" on the behaviour in multiple infarcted dementia (MID) rats, meanwhile to investigate the relationship between behaviour and apoptosis regulating genes-bcl-2 and bax in hippocampus and cortex region.Methods-. MID model was established by inverse injection of thrombus throughexternal carotid artery, the ability of spatial reference memory were determined by Morris Water Maze (MWM), apoptotic cells in hippocampus and cortex region was observed by TUNEL, expression of bcl-2 and bax genes were detected by immunohistochemistry(IHC)and in situ hybridization(ISH)technique and quantitative analysis was made by image pattern analysis.Result:MWM indicated that:1. In MID group, the ability of learning acquisition, memory retaining and relearning decreased obviously, course of learning and memory was delayed, edge and random pattern strategy was applied.2. In treatment group, the ability of learning acquisition, memory retaining and relearning increased markedly, course of learning and memory was shortened, tendency pattern strategy was applied.Index suggested that:1. In MID group, There existed multiple infarcted focuses in brain region . The amount of cell decreased in cortex, severe cell lost and cell arrangement disorder in hippocampus region. The number of apoptotic cell in hippocampus and cortex region was more than that of treatment group and control group (p<0.01), the expression of bcl-2 and bax genes enhanced, especially the expression of bax genes, bcl-2/bax descended.2. In treatment group, slight morphologic changes of cell, occasionally infarctedfocus, the number of local cell in cortex decreased, mild cell lost and cell arrangement in order relatively in hippocampus region. The number of apoptotic cell in hippocampus and cortex region was more than that of control group (p<0.01), and less than that of model group, the expression of bcl-2 and bax genes enhanced , especially the expression of bcl-2 genes, bcl-2/bax ascended.Conclusion: " YiQiTiaoXue, FuBenPeiYuan" therapy improved the symptoms ofdementia. The possible mechanism were presumed that acupuncture ascended the rate of bcl-2 and bax by increasing the expression of bcl-2 and suppressing bax expression, hence inhibited the apoptosis, furthermore improved the function of hippocampus and cortex, then advanced the ability of learning and memory.
