Xing-Hua-Yu Effecting On The Behavior Of Vascular Dementia Rats And The Expression Of Bcl-2and Bax Protein Of The Rat Hippocampus

Objective:This research is aim to further evaluate the capacity and part of possible functionmechanism of Xing-Hua-Yu’s improving memory ability by observing the influence ofXing-Hua-Yu effecting on the behavior of vascular dementia(VD) rats and the expression ofBcl-2and Bax protein of the rat hippocampus,which may provide experiment evidence forthe VD’s preventive treatment of this drug.Method:The VD model is established by the method of repeated ischemia-Reperfusion plusSodium Nitroprusside, the success replicate Model of VD rats were randomly grouped asfollows: model group,Xing-Hua-Yu A group,Xing-Hua-Yu B group and Jin-Na-Duogroup,in adition,pseudosurgery group.The next day,the model group and pseudeosurgerygroup are administrated saline solution by intraperitoneal injection respectively,all of themare given the same dose of5ml/kg,this procedure last21days.From the third weekends,weadopt the Morris water maze and the analysis system of dynamic image capture,recording allthe parameter by the coputer. The next day,after the experiment of Morris water maze,wedecapitate the rats and extract the brain tissue rapidly,and then do the HE staining to observethe pathology changes in the hippocampus area. Further more,we detect the expression ofBcl-2and Bax protein of the rat hippocampus by the immunohistochemistry.Results:1.Water maze experiment:From the fist day of the water maze train,there is nodifference between the psuedosurgery group and the others in the incubation period of escape(P>0.05). From the second day, the incubation period of escape of all of the groupssignificantly cut down, there is obvious difference between the model group and theothers(P<0.05).From the third day,the results are relatively stable,compaired with the modelgroup,the incubation period of escape of the Xing-Hua-Yu B group and psuedosurgery goupsignificantly cut down(P<0.01),and the Xing-Hua-Yu A group and the Jin-Na-Duo groupobviously cut down(P<0.05).The fourth to sixth day,there is significant difference betweenthe model group and the others(P<0.01),in addition,Jin-Na-Duo group and the Xing-Hua-Yu B group have obvious difference (P<0.05),but Jin-Na-Duo group and the Xing-Hua-Yu Agroup have no difference(P>0.05).2. Pathological changes:There is no obvious pathologychanges in the psuedosurgery group of the hippocampal CA1area shape of rats,And a largenumber of the rats hippocampal CA1area neuron of the model group are degeneration anddeath,presenting deepred staining,karyopyknosis,nucleus deep-staining. The Xing-Hua-YuA,B group and Jin-Na-Duo group compare with the model group,the hippocampal neuronbody are smaller,and their shape are approximately normal,and their degeneration and deathobviously alleviate,but only the hippocampal CA1area shape of rats in the Xing-Hua-Yu Bgroup are close to the pseudosurgery group.3. Bcl-2, Bax protein expression:There is asmall amount of Bcl-2,Bax positive cells expression in the hippocampal CA1area ofpseudosurgery group rats.The staining of Bcl-2,Bax positive cells of the model group aredeepened,and their number are obviously increased(P<0.01),and the change of Bax is moreobvious,the ratio of Bcl-2/Bax is lower than the pseudosurgery group(P<0.05). After thetherapy of Xing-Hua-Yu A,B and Ginkto,the number of Bax positive cells of ratshippocampal CA1area are decreased,especially in the Xing-Hua-Yu B group(P<0.01),whose ratio of Bcl-2/Bax is higher than the pseudosurgery group and model group(P<0.05).The number of Bcl-2positive cells in rats hippocampal CA1area of all of thegroup are increased(P<0.05),especially the Xing-Hua-Yu B group(P<0.01).Conclusion:1.The ginkgo flavonoids can significantly improve the cognitive ability and studymemory function of VD rats. With yhe same dose, Xing-Hua-Yu B has the best effect,Xing-Hua-Yu A correspond with Jin-Na-Duo group.2.Through the pathomorphology observation of hippocampal CA1area in brain tissue,we confirm that the ginkgo flavonoids can protect the neurocyte of hippocampal CA1area ofVD rats,and Xing-Hua-Yu B has the best effect.3.The results of immunohistochemistry confirm that the ginkgo flavonoids can preventand restrain the apoptosis of hippocampal neuron and relieve the damage of cerebral ischemiato the hippocampal neuron. Xing-Hua-Yu B has the strongest protective action.
