With the increase of aged population, Senile dementia gradually become the serious problem that greatly effect on the people's healthy and their life quality. Therefore, more and more scholars focus on the study of this disease. In the experiment, the animal models were established in Wistar rats by intraperitoneral injection of D-galactose for six weeks combined with ibotenic acid injection into bilateral nucleus basalis of Meynert. By applying electropuncture, the author has observed its effect on rats' memory and learning ability with rat shuttle devices, the expressions of bcl-2mRNA in rats' brain with In Situ Hybridization, the expressions of bcl-2 BDNF and bax proteins in rats' brain with SABC immunohistochemistry, apoptosis in hippocampus and cortex with Tunel, and the content of somatostatin (SS) in brain with radioommunoassay, and the content of acetylcholinesterase (AchE) and SOD in brain with colorimetric method. The results show that: acupuncture can significantly improve the memory ability of model rats; acupuncture can increase positive expression of bcl-2mRNA, bcl-2 and BDNF protein in the region of hippocampus and cortex and inhibit apoptosis to a certain extent; acupuncture can increase the content of SS in brain, which can nourish nerve cells and promote the recovery of damaged nerve cells; acupuncture can regulate the activity of AchE, SOD and infuence the content of Ach in brain , therefore can improve the memory and lerning ability of rats. The article explores further the mechanism of electropuncture treatment on impaired learning and memory and supplies the theoretical basis to the clinic.
