Irritable Bowel Syndrome (IBS), is a disease which exhibits intestinal dysfunction. Going with the economical developed and medicine improved, the incidence gradually rises. It has been become the most common functional disorder. Due to the classical diagnosis idea, it always depends on the positive finding of anatomical change and physiological abnormity. Because it is the typical functional disorder IBS has become the impeding of the research for IBS from further discovering the aetiological mechanisms and experimental animal model that is established for commonly acceptance So that it can not develop new drug to treat disease specifically. When the paper of research on IBS has been read, we realize that it is necessary to improve the ground-field in order to find the key of functional disease. We have noted that the disorder always went with the prescription of antibiotics. In the course of building animal model of IBS repeating test for it. We find the antibiotics might be the main cause of IBS that has been proved by using the drug to make intestinal bacteria flora to form cryptic growth cell (CGC). According to the clinical investigation of 300 cases belong to IBS, we find that almost everyone repeatedly had prescription of antibiotics. And nearly every patient suffer from bacterial CGCs intestinal tract. A lot of bacterial CGCs colonized mucosa. we have had studied the theory of bio- wave that indicate the CGCs as a stimulating factor. It stimulates intestinal mucosa owing to closely contact with the tissue, so that ultimately cause disorder. The design of double-effect of IBS aetiology has been supported by the positive results of population investigation ofantibiotics being an risk factor to IBS made by P.R.Maxwell. This thesissystematically analysed functional alteration in level of local tissue cell and molecule respectively. We found the interaction between CGCs and intestinal mucosal epithelia cells.And tried to establish experimental animal model of diarrhea-predominant IBS. The aim of the thesis is to establish the ground-field of the research on the mechanism of aetiological, and therapeutic effect of new drug for IBS.A number of techniques, such as microscopic-culture method, colonic electrical control activity(ECA)assay, fluorescence staining , molecular oscillation on cellular membrane scanned by AFM, and intracellular pH and Ca2+ assay and so on. Furthermore these techniques have been used to establish the animal model of IBS correlating with bacterial CGCs. So the criterion must be built for experimental diagnosis of IBS on different level established in series functional. The main contents and test results are as follows:1 . It is proved that a lot of substance such as antibiotics and some other chemical which are generally used, can cause the intestinal bacterial flora to transform into CGCs. In these condition, the new explored products for medicine such as antibiotics sterilized agents and so on appears stronger ability to cause CGCs. Whereas, the old products of antibiotics is not so stronger effect on CGCs formed. Bacterial CGCs exhibit different biological characteristics such as the form as filament, physiology as low metabolism, high-speed movement , multi-fashion of movements and abundant glycocalyx. The study of the characters of CGCs may help to realized the effects on aetiology of IBS.2. By means of colonization and stabilization in growth, bacterial CGCs is a permanence stimulus to affect the intestinal function. The correlation with the quality of CGCs ,course of illness, symptom of the patient and the relapse of IBS had been preliminarily determined.3. An experimental animal model of IBS had been established. The method is described as following. Cefuroxime was injected to the Wistarrats,weights 200-250 grams, liquid of pimiento was filled into rat's stomach by perfusion ,in a ratio of 1 :8 of weight of pimiento liquid vs rat weight, once a day, for three days .
