’The lung associated with the large intestine" which originated in the "Huangdi Neijing", is one of the important basic theories of Traditional Chinese Medicine. For thousands of years, people discussed and kept mining this theory deeply all the time, which makes the basic theory made new development from the theoretical connotation to clinical continuously, that play a very important role for guiding the Chinese Medicine clinic.In recent years, some scholars conducted in-depth of this theory, from literature and clinic studies, modern molecular biology, immunology, embryology, endocrinology, and constantly explored the mechanism of "The lung associated with the large intestine" from each level, and investigated the inner link between each kind of mechanism.The lung and the large intestine have a common structural basis. In physiology, the lung and the large intestine are linked by the regulatory effect of the nerve, hormone and body fluids, and they can also influence each other in the pathology. The theory of "The lung associated with the large intestine" has a biological basis:the lung and the large intestine in histogenesis have a homology in a certain extent, mucosal immunity may be the relationship of the lung and large intestine in effected substance basis.In clinic, according to the theory "The lung associated with the large intestine", the different treatments are formulated:there are"lung disease cure the bowel" "bowel disease cure the lung" and "lung and intestine are cured meanwhile"’, compared with old treatment, the effect is significantly improved. "The lung associated with the large intestine" have many new progress through the study of ancient and modern scholars, which made outstanding contributions to the guidance of the clinical Traditional Chinese Medicine and made a combination of Chinese traditional and Western Medicine.1 Research objectivesAround the classic theory of "The lung associated with the large intestine" this research summarizes the theoretical connotation and discusses the relationship between the lung and intestinal issue and related substances, in order to get the new material support about lung associated with the large intestine in the microcosmic, and enrich the connotation of "the lung and large intestine" further, it will supply the new methods and ideas for the modern study for Chinese Viscera-state Doctrine. In the first experiment, from the point of view of "bowel disease impact the lung" and "lung disease impact the intestinal", through the establishment of disease animal model about bowel disease model and pulmonary animal model,and observe the rats model in the general behavior and examine whether there is a certain synergetic recognition in lung and the large intestine in MYD88, IRAK and TRAF6 these LPS signal molecules explore the transfer node and its pathological transmission mechanism of "bowel disease impact the lung" and "lung disease impact the intestinal".In the second experiment, take the TCM theory of "The lung associated with the large intestine" as the guiding ideology, through the establishment of animal model of lung and bowel disease, selected two different time points (eighth days, sixteenth days) to observe the expression and correlation in different organs of related active substances such as SP, VIP, iNOS, CGRP in order to explore the pathological essence of "The lung associated with the large intestine".2 Research methods2.1 Literature ReviewThis paper applies one of the very important theories "The lung associated with the large intestine" from viscera correlation theory and the literature review methodology in the ancient clinical literature to record and collate, and using the theory of Traditional Chinese Medicine study on occurrence of methodology of Traditional Chinese Medicine to do the objectively and exactly interpretation about the lung and the large intestine.And conduct the research of the relevance theory about the lung and the large intestine from the angle of Western medicine: and explain the physiological, pathological relationship about the lung diseases and the bowel diseases, discuss the material basis of the theory, it not only enriches the connotation of theory, but also will be applied to clinical guidance rules.2.2 Experimental research2.2.1 Experimental study of the lung and the large intestine about MYD88, TRAK, TRAF6 of the LPS signal pathway45 Healthy female SD rats, the body weight of each rat is between 110 to120g, were randomly divided into five groups:normal group, food restriction group, limited water group, high oxygengroup and hypoxia group.Natural light, the temperature is between 24℃-26℃,the humidity is 40%-60%,The jejunum, ileum, colon and lung tissues of the five groups of rats were taken as the research object, test the content of each tissue in the five groups in MYD88, TRAK, and TRAF6 with Enzyme-Linked Immunosorbnent Assay (ELISA)2.2.2 Experiment Study of the correlation between the lung and the large intestine of neuropeptide substanceThe experiment used two groups of healthy female SD rats, the body weight of each rat is between 110 to 120g, and each group is 35 rats.35 rats of each group were divided into 5 groups:normal group, food restriction group, limited water group, high oxygengroup and hypoxia group.Natural light, the temperature is between 24℃-26℃,the humidity is 40%-60%, the lung, spleen, liver, bladder, kidney, serum, stomach, heart, jejunum, ileum, colon,rectum tissue in the five groups are taken and tested the content of CGRP、iNOS、SP and VIP with Enzyme-Linked Immunosorbnent Assay (ELISA) at two different time points (eighth days, sixteenth days).3 Results3.1 Results of first experimentThe jejunum:MYD88 levels of food restricted group were decreased than those in the normal group (p<0.05), MYD88 of high oxygen group increased compared with the food restricted group (p<0.05), MYD88 of high oxygen group increased compared with the water restricted group (p<0.05) also.The jejunum:IRAK level of high oxygen group increased than other group (p<0.05), IRAK level of hypoxia group increased compared with the water restriction group (p<0.05).The colon:TRAK6 level of limited water group, high oxygen group and hypoxia group decreased than the normal group (p<0.05), TRAK6 level of high oxygen group was lower than the limited food group(p<0.05), jejunum:TRAK6 level of high oxygen group are higher than normal group, food restriction group and limited water group (p<0.05).There are no statistically significant difference among other groups with multiple comparisons (p>0.05).3.2 Results of the second experimentThe average difference of the CGRP level in each organ of the five groups at different time points was not statistically significant.In the five groups, the iNOS level of food restricted group was higher than the water restriction group and hypoxia group in the bladder at sixteenth days; The SP level of food restricted group was higher than the normal group in the stomach at the eighth day; The VIP level of hypoxia group is higher than normal group, food restriction group, limited water group and hyperoxia group of spleen at the eighth day. There is no statistically significant difference among other groups with multiple comparisons.The linked organs in CGRP are:the lung and the heart(eighth days),the lungs and the jejunum(sixteenth days), the kidney and the the bladder (eighth days), the spleen and the liver (sixteenth days), the heart and the ileum (sixteenth days); The linked organs in iNOS are:the lung and bladder (sixteenth days), the heart and the rectum (sixteenth days); The linked organs in SP are:the lung and the liver (eighthdays and sixteenth days), the lung, the colon and the ileum(eighth days), the spleen and the stomach (eighth days), the heart and the ileum (eighth days); The linked organs in VIP are:the lung and the liver (eighth days), the heart and the colon (eighth days and sixteenth days).4 ConclusionsThe first experiment proved that TRAF6 and IRAK of the jejunum in pulmonary disease model (hyperoxia group) increased than the normal group, it provides experimental evidence for hypothesis of "the lung disease can lead to the bowel disease", and it can explain that there is a close relationship in the inflammatory process of the lung and the large intestine.In the second experiment, we can see in the four neuropeptides index, in CGRP and SP, the heart and the ileum associated; in VIP, the heart and the rectum are associated;so the connection between the heart and the intestinal is close, this is also a direction for future research. The lung is associated with the ileum and the colon in SP index, is associated with the jejunum in CGRP; is associated with the bladder on the iNOS.
