| Metabolic syndrome (MS) is a group of clinical syndrome, as the main clinical mani-festations includes insulin resistance (IR), the central obesity, diabetes or impaired glucose regulation, hypertension, abnormal lipid metabolism and so on. With the increasing of obese patients, the incidence of MS has reached epidemic proportions, which is recognized as a significant health problem according to human health.The central obesity which also known as abdominal obesity or visceral obesity, is refers to the main accumulation of fat in the abdomen or abdominal obesity. Excessive fat deposition in the visceral can cause the plasma free fatty acid (FFA) increased, promote gluconeo-genesis, make fat and inflammatory cytokines release increased, causing IR and abnormal glucose and lipid metabolism, leading to a variety of metabolic disorders. Consequently, how to control the excessive accumulation of visceral fat effectively and reversal metabolism disorder is one of the important means of prevention of metabolic syndrome from the source. Objective:The experimental study of acupuncture Daimai point on the MS rats and observe the body weight, abdominal circumference, food intake, metabolism of blood glucose and lipid regulating effect as well as the effects of PPARgamma and PPAR beta/delta gene and protein expression level in visceral adipose tissue.To study the effect of acupuncture Daimai point comprehensive therapeutic effect on rats with metabolic syndrome and related mechanisms.Method:The experiment is divided into three parts:(1) Establishment of rat model of metabolic syndrome:Feeding7weeks old SD rats with high fat and suger diet for12weeks, establish the metabolism syndrome model.75SD rats are randomly divided into3groups, the blank group contain15, given normal feed; while the model group contains45, given high fat and suger diet; the prevention group15, given high fat and suger diet either and bilateral acupuncture Daimai point, which giving on electroacu-puncture at the same time.Once a day and each time for20minutes.12weeks later, Selected to meet the increased abdominal girth,and the normal weight group increased more than20%, with IR or/and FBG increased, HDL-C decreased and hy-pertriglyceridemia, in line with the five of the two is the successful model.(2) The model rats are randomly divided into the model group and the treatment group. Then, the treatment group is given acupuncture treatment, the same as the prevention group and the duration of treatment is4weeks. During the experiment, dynamic detection based index of rats in each group, including Food intake, body weight, waist circumference, fasting plasma glucose (FBG), triglyceride(TG), total cholesterol(TC),the high density lipoprotein (HDL-C), the low density lipoprotein(LDL-C),serum insulin (FINS) and serum free fatty acid (FFA), visceral fat weight and visceral fat weight ratio, and calculation of HOMA-IR index and Lee’s index either. Then observe the effect of acupuncture pulse points syndromeon metabolic com-ponents.(3) After the acupuncture treatment, RT-PCR is used to detect PPAR gamma and PPAR beta/delta gene expression level in visceral adipose tissue and using western blot method to detect PPAR gamma and PPAR beta/delta protein expression level in visceral adipose tissue.To explore the possible mechanism for preventing and treating metabolic syndrome in the mole-cular level. Results:1The first experiment(1) Compares with the blank group, abdominal circumference, FBG, FINS, HOMA-IR, TG, TC, LDL-C levels of the model group are significantly higher than the blank group. There is significant difference between the two groups (P<0.05).(2) Compares with the model group, weight, FINS, HOMA-IR of the prevention group are lower than the model group, there is significant difference between the two groups (P<0.05); abdominal circumference, FBG, TC, TG and LDL-C are lower than the model group, HDL-C is higher than the model group, however, they have no signifinant difference between the two groups (P>0.05).(3) Compares with the blank group, abdominal circumference, TC, LDL-C are higher than the blank group, while the food intake is significant lower than it, there is statistical differences beween them (P<0.01); weight, FBG, TG, HDL-C are higher than the blank group,but they have no statistical differences (P>0.05).2The second experiment(1) Acupuncture Daimai point intervention for4weeks, the body weight, abdominal cir-cumference, food intake, Lee’s index, visceral fat weight, visceral fat weight ratio, FBG, FINS, HOMA-IR, TG, TC, LDL-C, FFA of the treatment group are obviously lower than the model group. There is significant difference between the two groups (P<0.05).(2) Acupuncture Daimai point intervention for4weeks, the body weight, abdominal circumference, length, food intake, Lee’s index, visceral fat weight, visceral fat weight ratio, FBG,FINS,HOMA-IR,TG,TC,LDL-C,FFA of the preventive group are obviously lower than the model group.There is significant difference between the two groups (P<0.05).(3) Acupuncture Daimai point intervention for4weeks, the food intake, FFA of the pre-vention group are lower than the blank group, there is statistical difference beween them (P< 0.05); Lee’s index, FINS, TC, visceral fat weight and visceral fat weight ratio are obviously higher than the blank group and there is statisticl difference (P<0.01); weght, FBG, HOMA-IR, TG,LDL-C are a little higher than the blank group, there is no significant difference, however (P>0.05).3The third experiment(1) Acupuncture Daimai point intervention for4weeks, PPAR gamma and PPARbeta/delta gene expression level in visceral adipose tissue of the treatment group are higher than the model group and the preventive group. There has significant differences (P<0.01).(2) Acupuncture Daimai point intervention for4weeks, PPARgamma and PPARbeta/delta gene expression level in visceral adipose tissue of the preventive group are higher than the model group but there has no significant differences (P>0.05).(3) Acupuncture Daimai point intervention for4weeks, PPARgamma and PPARbeta/delta gene expression level in visceral adipose tissue of the preventive group are lower than the blank group, there has statistics differences (P<0.05).(4) Acupuncture Daimai point intervention for4weeks, PPARgamma and PPARbeta/delta protein expression level in visceral adipose tissue of the treatment group are higher than the model group and the preventive group. There has significant differences (P<0.01).(5) Acupuncture Daimai point intervention for4weeks, PPARgamma and PPARbeta/delta protein expression level in visceral adipose tissue of the preventive group are a little higher than the model group, but there has no significant differences (P>0.05).(6) Acupuncture Daimai point intervention for4weeks, PPARgamma protein expression level in visceral adipose tissue of the preventive group is lower than the blank group, there has statistics differences (P<0.01); while the PPARbeta/delta protein expression level is a little lower than the blank group, there has no significant differences, however (P>0.05).Conclusions:1. Acupuncture Daimai point can reduce weight, abdominal circumference, food intake, FBG, FINS in MS rats and reduce the visceral fat, cholesterol and alleviate insulin resistance either.2. Early intervention of acupuncture Daimai point has significant effect in improving the preventive group rats body weight, abdominal circumference, visceral fat weight, visceral fat weight ratio, glucose and lipid metabolism and insulin resistance. Acupuncture Daimai point can prevent metabolic syndrome, one of the effect may be achieved by reducing weight, lipid and insulin resistance.3. Acupuncture Daimai can increase the relative expression of PPARy gene and protein in MS rats adipose tissue, which closely related to the activation of adiponectin, resistin in fatty tissues such as cytokines, promote the differentiation and apoptosis of adipocytes. And promote the expression of UCP1in mitochondrial brown fat cell membrane, who make the white adipose tissue browning.4. Acupuncture Daimai can increase the relative expression of PPARβ/δ gene and protein in MS rats adipose tissue, one of the possible way is that activated PPAR beta/δ effect on leptin and leptin receptor binding, and then inhibit the effection of NPY, which reduce food intake of rats and promote productivity, and inhibition of lipoprotein lipase in white adipose tissue (LPL) activity and promote the decomposition of lipid, and eventually reduce visceral fat accumulation.5. Acupuncture Daimai point has no significant effect in the relative expression of PPARγ and PPARβ/δ gene and protein in MS rats adipose tissue for the preventive group.We suggest that preventive effect of acupuncture Daimai point on MS may not through the regulation of PPARs signaling pathway on PPARy and PPARβ/δ expression. |
