By using current protocols of genetic engineering and protein engineering, a mutant encoding an analog ANP called sANPl with high efficiency and long half life ,and a mutant encoding analog IL-2 called as IL-2a were designed and cloned together to be a recombination plasmid,so as toget a novel bi-functional peptide.Such a construction was designed to get three kinds of peptide.It led to to utilize the products sufficiently from engineered host strain. It is an exploration at first time. The another aim of this research is that according to the function analysis on used genes, to design a targeting fusion protein ,so as to broaden the applied limits of such kind of therapeutic peptides.The research was divided into three parts.Part I: Construction and over expression of bi-functional peptide DFP1, whichcontains the three peptides including sANP1 and IL-2a in E.coli.Part II: Purification and characterization and sequencing of peptides DFP1 andsANP1.Part III: Fundamental investigation on pharmacological activities of DFP1 andsANP1.
